Abstract

Bakteri asam laktat merupakan jenis bakteri yang dapat menjaga keseimbangan mikroflora dalam saluran pencernaan ternak sepertikelinci (Oryctolagus cuniculus). Salah satu organ saluran pencernaan yang menjadi habitat bakteri asam laktat adalah rektum yang merupakan bagian dari usus besar paling akhir. Keberadaan bakteri asam laktat di dalam rektum sangat diperlukan untuk menghambat pertumbuhan bakteri patogen. Isolasi bakteri asam laktat dari rektum kelinci merupakan alternatif lain untuk mengisolasi bakteri asam laktat dari feses sehingga meminimalisir terjadinya kontaminasi. Penelitian ini bertujuan untukmengisolasi dan mengkarakterisasi bakteri asam laktatdari rektum kelinci. Sampel kelinci yang digunakan yaitu kelinci jantan berumur 35 hari sebanyak 3 ekor. Isolasi dilakukan dengan carabilasan dan kerokan mukosa rektum kelinci dan menggunakan media MRSA (deMan Rogosa Sharpe Agar) + CaCO3 1%. Karakterisasi bakteri dilakukan dengan uji morfologi koloni dan sel bakteri, uji fisiologi, dan uji biokimia yang meliputi uji katalase, oksidase, indol, reduksi nitrat, hidrolisis pati, dan fermentasi glukosa.Hasil isolasi diperolehbakteri asam laktat sampel bilasan dengan jumlah koloni rerata 10,44±12,65 x 104CFU/mL dan sampel kerokan rektum dengan jumlah koloni rerata3,38 ± 2,11 x 104CFU/mL. Terdapat 6 isolat bakteri asam laktat yang berhasil diisolasi, yangmenunjukkan hasil gram positif dan katalase negatif. Hasil uji biokimiamenunjukkan terdapat 2 isolat Lactobacillus plantarum, 2 isolat Lactococcus lactis, dan 2 isolat Lactobacillus acidophilus. Hasil uji potensi bakteri asam laktat sebagai kandidat probiotik menunjukkan bahwa bakteri asam laktatLactobacillus plantarum dan Lactococcus lactismempunyai kemampuan tumbuh yang tinggi pada media asam (pH 2) dan garam empedu (Oxgall 0,3 %), sehingga kedua isolat bakteri asam laktat hasil isolasi dari rektum kelinci (Oryctolagus cuniculus) memiliki potensi untuk dikembangkan sebagai probiotik.

English Abstract

Lactic acid bacteria is a type of bacteria that can maintain the balance of microflora in the digestive tract of animals such as rabbit (Oryctolagus cuniculus). One of digestive tract that becomes a habitat of lactic acid bacteria is the rectum which is part of the colon at the end. The fexistence of lactic acid bacteria in the rectum is needed to inhibit the growth of pathogenic bacteria. Isolation of lactic acid bacteria from the rectum of rabbits is another alternative to isolate lactic acid bacteria from feces in order to minimize contamination. This research was aimed to isolate and characterize the lactic acid bacteria from the rectum of rabbits. Rabbit samples that were used were buck of 35 days as much as 3 tails. Isolation was done by rinsing and mucosa scrapings of rabbitsrectum and using media of MRSA (deMan Rogosa Sharpe Agar) CaCO3 1%. Characterization of bacterial was done by colony morphology testing and bacterial cell, physiology tests, and biochemical tests that included catalase test, oxidase, indole, nitrate reduction, starch hydrolysis, and fermentation of glucose. The isolation result was obtained by the lactic acid bacteria of rinse samples by the number of colonies were average of 10.44±12.65 x 104CFU/mL and rectal scrapings samples with the colony numberwere3.38 ± 2.11 x 104CFU/mL. There were 6 isolates of lactic acid bacteria that could be isolated, which showed the results of positive gram and negative catalase. The results of biochemical tests showed that there were two isolates of Lactobacillus plantarum, 2 isolates of Lactococcus lactis, and 2 isolates of Lactobacillus acidophilus. The result of the potential of lactic acid bacteria as probiotic candidates showed that the lactic acid bacteria ofLactobacillus plantarum and Lactococcus lactishad to grow a high ability in an acidic media (pH 2) and bile salts (Oxgall 0.3%), so the two of isolates of lactic acid bacteria of the isolation result from rabbit’s rectum (Oryctolagus cuniculus) had the potential to be developed as probiotics.
