Abstract
Malaria merupakan penyakit yang bisa menimbulkan kematian berisiko tinggi di Indonesia. Penelitian ini dilakukan untuk membuktikan bahwa isolat senyawa flavonoid dari fraksi etil asetat daun Alectryon serratus memiliki aktivitas antimalaria dan setara dengan klorokuin. Metode penelitian berupa eksperimental laboratorium secara in vitro dengan mengukur nilai persen parasitemia, nilai persen hambatan, efek antimalarial (IC50). Isolat flavonoid yang digunakan berupa isolat A.1 dan A.2 (Kaempferol-3-O-glikosida). Pengukuran dilakukan secara duplo, serial kosentrasi 10 ug/mL, 1 ug/mL, 0,1 ug/mL, 0,01 ug/mL dan 0,001 ug/mL. Hasil uji korelasi Pearson antara dosis isolat A.1 dengan persen parasitemia berhubungan negatif (R=-0,601, p=0,066), uji Pearson antara dosis isolat A.1 dengan persen hambatan pertumbuhan parasit memiliki hubungan positif (R=0,601, p=0,066). Hasil uji korelasi Pearson antara dosis isolat A.2 dengan persen parasitemia berhubungan negatif (R=-0,629, p=0,052), uji Pearson antara dosis isolat A.2 dengan persen hambatan pertumbuhan parasit memiliki hubungan positif (R=0,629, p=0,051). IC50 isolat A.1 sebesar 0,011 ug/mL, IC50 isolat A.2 sebesar 0,010 dan IC50 klorokuin sebesar 0,001 ug/mL. Berdasarkan hasil tersebut disimpulkan bahwa isolat senyawa flavonoid dari fraksi etil asetat daun Alecryon serratus memiliki aktivitas antimalaria.
English Abstract
Malaria is an infection disease with high mortality number in Indonesia. This study was conducted to determine antimalarial activity of flavonoid compound isolated from the ethyl acetate fraction of Alectryon serratus leaves and equivalent with Klorokuin. The research method was experimental laboratory in vitro by measuring parasitemia percentage, growth inhibition percentage and antimalarial effect (IC50). Falvonoid isolated to use two compound. There were isolate A.1 and isolate A.2 (Kaempferol-3-O-glycosides). Measurements were performed by duplo, serial concentration of 10 ug/mL, 1 ug/mL, 0.1 ug/mL, 0.01 ug/mL and 0.001 ug/mL. Pearson test results correlation between dose isolate A.1 with percentage of parasitemia negative correlation (R=-0,601, p=0,066), Pearson test result correlation between dose isolate A.1 with percentage of parasite growth inhibition have positive relation (R=0,601, p=0,066). Pearson test results correlation between dose isolate A.2 with percentage of parasitemia negative correlation (R=-0,629, p=0,052) and Pearson test result correlation between dose isolate A.2 with percentage of parasite growth inhibition have positive relation (R=0,629, p=0,051). The result show IC50 of isolate A.1 was 0,011 ug/mL, IC50 isolate A.2 was 0,010 ug/mL and IC50 klorokuin was 0,001 ug/mL. It was concluded that the flavonoid compound isolated from ethyl acetate fraction of Alectryon serratus had antimalarial activity.
