Abstract
Kanker payudara merupakan suatu proses pembelahan sel pada jaringan penyusun payudara yang terjadi secara abnormal. Obat sintetik yang digunakan untuk pengobatan kanker payudara memberikan efek samping yang cukup berbahaya. Buah lemon memiliki kandungan senyawa yang bersifat antitumor, antiinflamasi, dan antioksidan. Tujuan dari penelitian ini adalah mengetahui pengaruh ekstrak buah lemon (Citrus limon) terhadap profil sel T CD4+CD25+ dan sel T CD4+CD25+ CD62L+ pada mencit betina (Mus musculus) model kanker payudara induksi DMBA. Mencit dibagi menjadi empat kelompok perlakuan dengan empat ulangan, yaitu Normal, DMBA, DMBA+Ekstrak Lemon 50 mg/kg BB, dan DMBA+Ekstrak Lemon 200 mg/kg BB. Mencit diinjeksi DMBA secara subkutan dengan dosis 0.015 mg/kg BB agar mengalami kanker payudara. DMBA diinjeksikan sebanyak satu kali dalam seminggu selama enam minggu. Kemudian, mencit diberi ekstrak buah lemon melalui oral, setiap hari selama 14 hari. Sel-sel imunokompeten diisolasi dari organ limpa kemudian dilakukan pewarnaan sel menggunakan antibodi dan dianalisis flow cytometry. Setelah itu, data hasil flow cytometry dianalisis secara statistik dengan uji One-way ANOVA pada signifikasi 5% menggunkan program SPSS versi 16.0 for windows. Hasil menunjukkan bahwa pada mencit kanker payudara, rata-rata jumlah relatif sel T CD4+CD25+ mengalami peningkatan dan sel T CD4+CD25+ CD62L+ mengalami penurunan, setelah diberi ekstrak buah lemon, ratarata jumlah relatif sel T CD4+CD25+ mengalami penurunan dan sel T CD4+CD25+CD62L+ mengalami peningkatan. Ekstrak buah lemon mengandung senyawa flavonoid yang berperan sebagai immunodulator.
English Abstract
Breast cancer is a process of cell growth in breast tissue composition that occurs abnormally. Synthetic drugs used for the treatment of breast cancer provide a fairly dangerous side effects. Lemon fruit contains antitumor, anti-inflammatory, and antioxidant. The purpose of this research is to know the effect of lemon (Citrus limon) extract on T CD4+CD25+ and T CD4+CD25+CD62L+ cells profile on female mice (Mus musculus) model of DMBA induction breast cancer. Mice were divided into four groups with four replications, are Normal, DMBA, DMBA+Lemon Extract 50 mg/kg BW, and DMBA+Lemon Extract 200 mg/kg BW. Mice injected DMBA subcutaneously with a dose of 0.015 mg/kg BW to have breast cancer. DMBA is injected once a week for six weeks. Then, the mice were treated with lemon extract through oral, daily for 14 days. Immunocompetent cells were isolated from the spleen organ and then stained cells using antibodies and analyzed flow cytometry. After that, flow cytometry data were analyzed statistically with One-Way ANOVA and Post Hoc Tukey HSD test on 5% significance using SPSS version 16.0 for windows. The results showed that in breast cancer mice, the mean relative number of T CD4+CD25+ cells is increased and T CD4+CD25+CD62L+ cells is decreased, after treatment with lemon extract, the mean relative number of T CD4+CD25+ cells is decreased and T CD4+CD25+ CD62L+ cells is increased. Lemon extract contains flavonoid that act as immunodulators.
