Abstract
Kanker merupakan penyakit penyebab kematian yang tinggi. Respon sistem imun yang dapat menginisiasi sel kanker adalah inflamasi. Buah lemon diyakini memiliki efek antiinflamasi untuk setiap tipe kanker dengan efek samping yang rendah. Tujuan dari penelitian ini adalah untuk mengetahui efek lemon (Citrus limon) terhadap profil sel T CD4, TNF-α, dan IFN-ɣ pada mencit model kanker payudara injeksi DMBA. Tahap awal yang dilakukan adalah dengan mempersiapkan hewan coba sebanyak 16 ekor betina ditempatkan dalam 4 perlakuan yang berbeda (Normal, DMBA, DL-50, DL-200). Bahan yang digunakan adalah ekstrak daging buah lemon dengan metode freeze dryer ditambahkan pelarut akuades. Mencit normal tidak diberi perlakuan sedangkan model kanker payudara diinjeksi dengan DMBA 6 kali secara subkutan. Setelah mencit kanker, kelompok mencit DMBA tidak diberikan perlakuan sebagai kontrol negatif sedangkan kelompok perlakuan diberi ekstrak lemon sesuai dosis masing-masing (DL-50 mg/kg BB dan DL-200 mg/kg BB) setiap hari selama 2 minggu. Setelah tahap perlakuan selesai dilakukan isolasi organ limpa, pewarnaan sel menggunakan antibodi dan analisis flowcytometry. Hasil data yang diperoleh dilakukan analisis statistika menggunakan uji one way ANOVA dengan nilai signifikansi 0,05. Hasil menunjukkan bahwa DMBA dapat menyebabkan kenaikan inflamasi berkaitan dengan peningkatan profil jumlah relatif sel T CD4+ yang mensekresi sitokin proinflamasi (TNF-α+ dan IFN-ɣ+) pemberian ekstrak lemon pada masing-masing dosis yaitu 50mg/kg BB dan 200mg/kg BB mencit selama 2 minggu perlakuan mampu menurunkan jumlah relatif sitokin proinflamasi.
English Abstract
Inflammation is the one of immune system response that can initiate cancer cell. Lemon is widely used because have antiinflamation effect for ever type of cancer with low side effect. The aim of this research is to know the effect of lemon (Citrus limon) on mice breast cancer based on T cell CD4, TNF-α, IFN-ɣ profiles. First step is preparing 16 female mice which are placed in 4 different treatment (Normal, DMBA, DL-50,and DL-200). The material used is fruit of lemon extracted using freeze dryer solvent by aquades. Mice are injected with DMBA 6 times on subcutaneous injection except normal mice (N) group. After cancer treatment, group of lemon treatment given each appropriated dose lemon extract (DL-50 mg/kg BB and DL- 200 mg/kg BB) every day for 2 weeks, group of DMBA given nothing as negative control. After that begin to spleen isolation, staining cell with antibody, and analysis using flowcytometry. Obatained data from result analyzed using one way test ANOVA with a value of 0,05 significance. The result that DMBA can caused increase inflammation related with relative number of CD4 profile that are secreting proinflammatory cytokines (TNF-α and IFN-ɣ). After mice gave extract lemon as treatment for 2 weeks that can decrease number of relative proinflammatory chytokines.
