Abstract
Penelitian mengenai perbandingan enzim lipase dari Rhizomucor miehei dan Candida antarctica recombined Aspergillus oryzae untuk mengkatalisis reaksi asetilasi senyawa mentol dengan perbedaan aktivitas dari enzim tersebut. Reaksi dilakukan pada kondisi temperatur 50oC menggunakan sumber asetil asetat anhidrid dalam pelarut n-heksan pada berbagai variasi waktu. Hasil yang diperoleh yaitu reaksi asetilasi mentol menggunakan enzim lipase dari Candida antarctica recombined Aspergillus oryzae dan Rhizomucor miehei dengan unit aktivitas yang berbeda tidak memiliki perbedaan yang signifikan. Perbedaan hasil produk akhir yaitu mentil asetat dari kedua enzim mengalami perubahan seiring dari waktu ke waktu menghasilkan perbedaan hasil pada jam ke – 24. Penggunaan lipase dari Candida antarctica recombined Aspergillus oryzae mempunyai selektivitas yang lebih tinggi (96,93%) dalam membentuk mentil asetat dibanding lipase dari Rhizomucor miehei dengan mempunyai kemampuan mengkonversi sebesar 77,42%.
English Abstract
Research on the comparison of lipase enzymes from Rhizomucor miehei and Candida antarctica in Aspergillus oryzae recombination to catalyze the reaction. The reaction is carried out at a temperature of 50 ° C using an anatide acetyl source in a n-hexane solvent at various time variations. The resulting result is the reaction of menthol acetylation using lipase enzyme from Candida antarctica recombination of Aspergillus oryzae and Rhizomucor miehei with different activity unit has no significant difference. The difference in end product yields is menthyl acetate of both things which can change the results at 24 hours. The use of lipase from Candida antarctica recombined Aspergillus oryzae has higher selectivity (96,93%) in forming mentyl acetate than lipase from Rhizomucor miehei with conversion ability 77,42%.
