Abstract

Studi mengenai reaksi asilasi senyawa l-menthol dengan katalis enzim lipase dari Candida antarctica recombined Aspergilus oryzae telah dilakukan. Pada penelitian ini reaksi asilasi dilakukan melalui uji in vitro dengan menggunakan variasi asil, yaitu asam asetat anhidrid dan vinil asetat. Reaksi dilakukan pada temperatur 50oC dengan pelarut n-heksan pada berbagai variasi waktu. Hasil yang diperoleh enzim lipase Candida antarctica recombined Aspergilus oryzae mampu mengkatalis reaksi asilasi l-menthol menjadi l-menthil asetat. Dari hasil penelitian, %l-menthol, selektivitas, dan konversi meningkat seiring dengan meningkatnya temperatur dan lama reaksi. Hasil terbaik reaksi diperoleh pada reaksi asilasi menggunakan asam asetat anhidrid sebagai sumber asil pada jam ke-24. %l-menthil asetat tertinggi yaitu menggunakan sumber asil asam asetat anhidrit pada jam ke-24 dengan nilai sebesar 58,45%. Hasil tersebut bersesuaian dengan hasil dari uji in silico. Dimana efektivitas masing-masing sumber asil tersebut dihitung melalui nilai Kd, dengan Kd dari asetat anhidrid lebih besar dibandingkan Kd vinil asetat.

English Abstract

The study of acylation reaction of l-menthol with lipase from Candida antarctica recombined Aspergilus oryzae has been performed. In this study, the acylation reaction was done through in vitro using acyl variation, that is acetic acid anhydride and vinyl acetate. The reaction is carried out at the temperature of 50 ° C with n-hexane solvent at various time. The results is lipase from Candida antarctica recombined Aspergilus oryzae were able to catalyze the l-menthol acylation reaction to l-menthyl acetate. From the results, %l-menthol, selectivity, and conversion increases with increasing temperature and reaction time. The best result of the reaction is in the acylation reaction using acetic acid anhydride as an acyl source at 24 hours. % L-menthil acetate using acetic acid anhydride at 24 hours is 58.45%. These results correspond to the results of the in silico. The effectiveness of each source of acyl is calculated through the value of Kd, which Kd of anhydride acetate is greater than Kd of vinyl acetate.
