Abstract
Asma merupakan penyakit kronik pada saluran pernafasan yang banyak dijumpai pada hewan dan manusia. Asma dihubungkan dengan hiperresponsif bronkus, hipersekresi mukus dan gejala pernapasan yang bersifat reversibel. Salah satu sel yang diketahui berperan besar dalam patogenesis asma adalah eosinophil yang berperan dalam merusak epitel saluran napas dan menyebabkan peradangan. Gejala asma dapat dipicu oleh ovalbumin dan dapat diperparah oleh infeksi rongga mulut akibat Lipopolisakarida (LPS) dari bakteri Gram negatif. Tomat mengandung antioksidan yang berguna untuk menangkal radikal bebas. Whey kefir merupakan produk susu fermentasi yang kaya akan senyawa antimikroba diantaranya asam organik, peptida dan eksopolisakarida. Tujuan penelitian ini untuk mengetahui potensi terapi kombinasi tomat (Solanum lypopersicum l) dan whey kefir terhadap kadar Malondialdehida (MDA) dan jumlah eosinofil pada hewan tikus model asma. Tikus model asma diinduksi ovalbumin diinjeksikan secara intraperitoneal 10 μg dengan 1,5 mg AlOH3 dalam 200 μL PBS (phosphate buffer saline) sebanyak 3 kali, induksi yang pertama dan kedua diinjeksi intraperitoneal dan ketiga diinhalasi dengan nebulizer dan dipapar LPS dari bakteri Phorphyromonas gingivalis secara intrasulkuler dengan dosis 1 μg. Penelitian ini menggunakan lima kelompok perlakuan, yaitu kontrol negatif, kontrol positif, perlakuan 1, 2 dan 3 diterapi kombinasi tomat (Solanum lypopersicum l) dan whey kefir dengan dosis yang berbeda perlakuan 1ml/200kgBB, 1,5ml/200KgBB dan 2ml/200KgBB. Kadar MDA diukur menggunakan metode Thiobarbaturic Acid (TBA) dan perhitungan jumlah eosinophil dengan ABX Micros 60. Data yang diperoleh berapa data kuantitatif uji MDA dan jumlah eosnofil selanjutnya data dianalisa menggunakan uji analisis ragam ANOVA dan uji lanjutan Beda Nyata Jujur (BNJ) α = 5%. Hasil penelitian didapatkan bahwa pemberian whey kefir dan tomat mampu menurunkan kadar MDA dan menurunkan jumlah eosinofil. Dosis terapi terbaik yaitu 1.5 ml/200g BB. Kesimpulan penelitian ini yaitu kombinasi whey kefir dan tomat dapat digunakan sebagai terapi tikus model asma.
English Abstract
Asthma is a chronic disease of the respiratory tract that are often found in animals and humans. Asthma is associated with bronchial hyperresponsiveness, hypersecretion of mucus and respiratory symptoms are reversible. One cell is known to play as a major role in the pathogenesis of asthma is eosinophil that play a role in airway epithelial damage and cause inflammation. Asthma symptoms can triggered by ovalbumin and can be aggravated by oral infections due to lipopolysaccharide (LPS) from Gram-negative bacterias. Tomatoes have antioxidants that are useful to prevent free radicals. Whey kefir is a fermented milk product with a lot of antimicrobial compounds include organic acids, peptides and exopolysaccharide. The purpose this study to determine the potential of combination therapy tomato (Solanum lypopersicum l) and whey kefir against Malondialdehida levels (MDA) and the number of eosinophils in rat as an animal models of asthma. Rat model of ovalbumin-induced asthma were injected intraperitoneally 10 mg to 1.5 mg AlOH3 in 200 mL of PBS (phosphate buffer saline) 3 times, the induction of the first and the second and third were injected intraperitoneally with a nebulizer and is exposed to inhaled LPS from Phorphyromonas gingivalis bacteria in intrasulkuler with a dose of 1 mg. This study uses five treatment groups, is the negative control, positive control, treatment 1, 2 and 3 treated with a combination of tomatoes (Solanum lypopersicum l) and kefir whey treatment with different doses of 1ml / 200kgBB, 1,5ml / 200KgBB and 2ml / 200KgBB. MDA levels were measured using the Thiobarbaturic Acid (TBA) method and the calculating the number of eosinophil with ABX Micros 60. Data obtained quantitative data turned away MDA test and subsequent eosnofil amount of data analyzed using analysis of variance ANOVA test and advanced test Honestly Significant Difference (HSD) α = 5 %. The results showed that whey kefir and tomato were able to decrease MDA levels and decrease the amount of eosinophils. The best therapy dose is 1.5 ml / 200 g BW. The conclusion of this research is the combination of whey kefir and tomato can be used as therapy in rats as animal model of asthma.
