Abstract

Penyebab kematian dan kesakitan utama pada penderita diabetes melitus (DM) adalah penyakit kardiovaskuler seperti coronary artery disease. Salah satu pengobatan untuk mecegah coronary artery disease adalah konsumsi antidiabetik. Antosianin dari ekstrak ubi jalar ungu memiliki kemampuan antiabetik. Penelitian ini bertujuan untuk membuktikan pengaruh pemberian Antosianin terhadap ketebalan tunica intima hingga adventitia cabang arteri koronaria tikus (rattus norvegicus) Wistar yang diinduksi streptozotocin (STZ) dan diet tinggi lemak. Studi eksperimental ini menggunakan post test only controlled group design. Sampel dibagi menjadi 5 kelompok yang masing-masing terdiri dari 4 ekor tikus, yaitu kelompook control negatif (normal), kontrol positif (induksi STZ dan diet tinggi lemak), kelompok perlakuan 1 (induksi STZ dan diet tinggi lemak + Antosianin dosis 10mg/kgBB/hari), kelompok perlakuan 2 (induksi STZ dan diet tinggi lemak + Antosianin dosis 20mg/kgBB/hari), kelompok perlakuan 3 (induksi STZ dan diet tinggi lemak + Antosianin dosis 80mg/kgBB/hari). Variabel yang diukur pada penelitian ini adalah ketebalan tunika intima hingga adventitia cabang arteri koronaria tikus. Hasil deskriptif dari pengukuran rerata ketebalan cabang arteri koronaria tikus menunjukan adanya kecenderungan terjadi penurunan ketebalan pembuluh darah seiring dengan ditambahnya dosis Antosianin, namun uji one-way ANOVA didapatkan nilai signifikansi 0,331 yang berarti tidak signifikan. Kesimpulan dari penelitian ini adalah pengaruh pemberian Antosianin terhadap ketebalan tunika intima hingga adventitia cabang arteri koronaria tikus model diabetes melitus belum dapat dibuktikan.

English Abstract

The main cause of morbidity and mortality in patient with diabetes mellitus (DM) is cardiovascular disease, especially coronary artery disease. One of available treatment to prevent the coronary artery disease in DM is through the consumption of antidiabetic. Anthocyanin from the Extract of Purple Sweet Potato has the antidiabetic effect. This research was aimed to prove the effect Anthocyanin to decrease the thickness of the branch of coronary artery tunica intima to tunica adventitia of Wistar rats (Rattus norvegicus) induced by streptozotocin (STZ) and high fat diet. This experimental research used post test only controlled group design. The samples were divided into 5 groups, each group consisted of 4 rats, the negative control group (normal), positive control group (induction of STZ and high-fat diet), dose 1 group (induction of STZ and high-fat diet + Anthocyanin 10mg/kg/day), dose 2 group (induction of STZ and high-fat diet + Anthocyanin 20mg/kg/day), dose 3 group (induction of STZ and high-fat diet + Anthocyanin 80mg/kg/day). The measured variable in this research was the thickness of the branch of coronary artery’s tunica intima to tunica adventitia. The result from mean descriptive test showed a tebdency to decrease the thickness of blood vessels along with the added dose of Antosianin, but One-way ANOVA was used to analyze various dose that showed significance score 0.331 which means not statistically significance. The conclusion of this research is that Anthocyanin administration affect the thickness of the branch of coronary artery’s tunica intima to tunica adventitia in diabetic induced rat has not been proven.
