Abstract

Infeksi Candida albicans dapat berupa kandidiasis, dimana infeksi ini banyak terjadi pada wanita diseluruh dunia. Obat-obatan antifungi memiliki efek samping yang merugikan seperti gangguan alat cerna, iritasi, reaksi alergi, gangguan endokrin, serta memicu munculnya resistensi. Daun wungu (Graptophyllum pictum) memiliki kandungan aktif yang diduga bermanfaat sebagai antifungi antara lain flavonoid, saponin, tanin, alkaloid, dan minyak atsiri. Tujuan dilaksanakannya penelitian ini adalah untuk mengetahui bahwa esktrak etanol daun wungu memiliki efektivitas sebagai antifungi terhadap Candida albicans secara in vitro. Penelitian ini merupakan penelitian eksperimental laboratorik dengan menggunakan metode dilusi agar untuk menentukan Kadar Hambat Minimal (KHM). Sampel yang digunakan dalam penelitian ini adalah jamur Candida albicans yang diperoleh dari Laboratorium Mikrobiologi Universitas Brawijaya, Malang, Jawa Timur. Konsentrasi ekstrak yang digunakan dalam penelitian ini adalah 0%, 15%, 20%, 25%, 30%, dan 35% dengan empat kali pengulangan. Hasil uji analisis Kruskal Wallis didapatkan perbedaan yang signifikan (p=0.000), kemudian uji lanjutan dengan Mann Whitney. Uji Korelasi Spearman menunjukkan terdapat hubungan terbalik antara kenaikan konsentrasi ekstrak etanol daun wungu dengan pengurangan jumlah koloni Candida albicans (p= -935). Kesimpulan dari penelitian ini adalah ekstrak etanol daun wungu (Graptophyllum pictum) efektif dalam menghambat pertumbuhan koloni Candida albicans dengan Kadar Hambat Minimal (KHM) pada konsentrasi 25%.

English Abstract

Candida albicans infections may emerge as candidiasis, in which infections are more likely to occur in women throughout the world. Anti-fungi medications possess harmful side effects such as gastrointestinal irritation, irritation, allergic reactions, endocrine disorders, and the chance of resistance. Wungu leaves (Graptophyllum pictum) has active compounds that were suspected to act as anti-fungi, such as flavonoid, saponin, tanin, alkaloid, and essential oil. The objectives of this research are to figure whether the ethanol extract from Graptophyllum pictum’s leaves may act as anti-fungi toward Candida albicans in vitro. This research is laboratory experiment using dilution method to determine the Minimal Inhibitory Concentration (MIC). The sample in this research were Candida albicans fungi obtained from Brawijaya University Microbiology Laboratory, Malang, East Java. Extract concentrates that were used in this research were 0%, 15%, 20%, 25%, 30%, and 35%, each with four repetitions. Through the examination results on Kruskal Wallis analysis, a significant difference was obtained (p=0.000), then further examined with Mann Whitney. Spearman’s correlation examination showed inversed correlation between the increase of Graptophyllum pictum’s leaves ethanol extract concentration rate and the decrease of Candida albicans colony (p= -935). The conclusion of this research is that Graptophyllum pictum’s leaves ethanol extract may resist the growth of Candida albicans colony effectively with Minimal Inhibitory Concentration (MIC) of 25%.
