Abstract

Traumatic Brain Injury atau Cedera Otak merupakan suatu gangguan traumatik yang bersifat mekanis dari otak. Kasus cedera otak pada dunia veteriner paling banyak disebabkan oleh kecelakaan (automobile accident), jatuh dari ketinggian, tendangan, gigitan hewan lain dan crushing injuries. Tujuan dari penelitian ini adalah untuk mengetahui ekspresi Interleukin 6 dan gambaran Histopatologi Cerebrum pada hewan coba tikus (Rattus norvegicus) model Traumatic Brain Injury dengan metode induksi Weight Drop Injury. Hewan coba yang digunakan dalam penelitian ini adalah tikus (Rattus norvegicus) berumur 8- 12 minggu dengan berat badan antara 300 - 450 gram. Tikus dibagi menjadi 2 kelompok yaitu kelompok kontrol dan kelompok perlakuan. Induksi Traumatic Brain Injury menggunakan metode Weight Drop Injury (WDI) atau penjatuhan beban dengan berat 50 g dari ketinggian 1,5 m pada medial kepala tikus. Parameter yang diamati adalah ekspresi Interleukin 6 dengan metode pewarnaan imunohistokimia dan gambaran histopatologi cerebrum dengan pewarnaan Hematoksilin-Eosin (HE). Data kuantitatif berupa data presentase area ekspresi Interleukin 6 dilakukan analisa statistik menggunakan uji T tidak berpasangan (Independent T student test) dengan α = 5% untuk mengetahui hasil perlakuan. Sedangkan data kualitatif berupa gambaran histopatologi cerebrum dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa pada kondisi Traumatic Brain Injury terjadi peningkatan Ekspresi Interleukin 6 yang signifikan sebanyak 33,988%. Gambaran Histopatologi Cerebrum tikus menunjukkan adanya perubahan histopatologi yang ditandai dengan adanya nekrosis, sel radang dan pendarahan. Kesimpulan dari penelitian ini adalah Traumatic Brain Injury dapat meningkatkan ekspresi Interleukin 6 dan menyebabkan nekrosis pada cerebrum.

English Abstract

Traumatic Brain Injury is a traumatic disorders of the brain that is mechanical causes. Traumatic Brain Injury is also frequently encountered in veterinary practice as a result of automobile accidents, falls, assaults, bites, and crushing injuries. The aim of this study was to determine the expression of Interleucine 6 and picture Histopathology cerebrum in rats (Rattus norvegicus) model of Traumatic Brain Injury with induction Weight Drop Injury method. Experimental animals used in this study was a rat (Rattus norvegicus) 8-12 weeks old weighing between 300 - 450 grams. Induction of Traumatic Brain Injury using Weight Drop Injury (WDI) methods or imposition of burdens weighing 50 g from a height of 1.5 m on the medial head of rats. Rats were divided into 2 groups: control group and treatment group. Staining methods used to see the expression of Interleucine 6 is the method of immunohistochemistry. Histopathology image of cerebrum was observed by hematoxylin-eosin staining (HE). This research data in the form of quantitative and qualitative data. Interleucine 6 expression is obtained from the quantitative data that were then tested using Independent T student test (α = 5%) to determine treatment outcome. The qualitative data of the brain cortex histopathology image were analyzed descriptively. The results of this study showed that the condition of the Traumatic Brain Injury can increase the expression of interleucine 6 significantly as much as 33,988%. The cerebrum of rats has histopathological changes and marked by the presence of necrosis, inflammatory cells and haemorrhage. The conclusion of this study is Traumatic Brain Injury can increase expression of Interleucine 6 and causes necrosis of cerebrum.
