Abstract

Rhodamin B merupakan zat warna sintetik yang dilarang digunakan dalam produk pangan karena menimbulkan iritasi mukosa saluran pencernaan, saluran pernafasan dan kulit. Bawang putih mengandung antioksidan berupa alisin berfungsi menurunkan serta mencegah timbulnya radikal bebas. Penelitian bertujuan mengetahui pengaruh pemberian air perasan bawang putih (Allium sativum) terhadap aktivitas protease dan perubahan gambaran histopatologi jejunum tikus (Rattus norvegicus) yang diberi rhodamin B melalui pakan. Penelitian menggunakan tikus jantan strain Wistar dibagi menjadi kelompok kontrol negatif, kelompok kontrol positif diberi pakan mengandung rhodamin B dengan konsentrasi 600 ppm, kelompok terapi preventif P1, P2, P3 diberikan air perasan bawang putih dengan volume pemberian 0,5mL/200gBB, 1,0mL/200gBB, 1,5mL/200gBB dengan konsentrasi air perasan bawang putih 25% setelah dipapar rhodamin B melalui pakan selama 14 hari. Aktivitas protease diukur dengan metode spektrofotometri dan histopatologi jejunum diamati menggunakan pewarnaan HE. Data aktivitas protease dianalisa secara kuantitatif menggunakan ANOVA, sedangkan histopatologi jejunum dianalisa secara deskriptif. Hasil penelitian menunjukkan air perasan bawang putih memberikan pengaruh nyata (p<0,05) terhadap penurunan aktivitas enzim protease dan histopatologi jejunum menunjukkan berkurangnya kerusakan sel epitel yang ruptur, infiltrasi sel radang, dan sel goblet yang hipertrofi. Air perasan bawang putih 0,5 ml/200gBB dengan konsentrasi air perasan bawang putih 25% merupakan volume efektif dalam menurunkan aktivitas enzim protease sebesar 43% dan mencegah kerusakan gambaran histopatologi jejunum tikus (Rattus norvegicus).

English Abstract

Rhodamin B is a prohibited synthetic dyestuff used in food products because it irritates the digestive tract mucosa, respiratory tract and skin. Garlic contains antioxidant allicin and serves to reduce and prevent the occurrence of free radicals. The aim of this research is to investigate the effect of garlic juice (Allium sativum) on the protease activity and change of histopathologic image of rat’s jejunum (Rattus norvegicus) induced by rhodamine B through feed. The experiment used male rat wistar strain divided into negative control group, positive control group fed rhodamine B with concentration of 600 ppm, preventive therapy group P1, P2, P3 given garlic juice with a volume of 0.5mL/200gBB, 1.0mL/200gBB, 1.5mL/200gBB with 25% garlic juice concentration after exposure to rhodamine B through feed for 14 days. Protease activity measured by spectrophotometric method and histopathology of jejunum was observed use HE staining. Protease activity data were analyzed quantitatively used ANOVA, while jejunum histopathology was analyzed descriptively. The results showed that garlic juice had significant effect (p <0,05) on the decrease of protease enzyme activity and histopathology of jejunal organ showed decrease of ruptured epithelial cell damage, inflammatory cell infiltration, and hypertrophy goblet cells. The volume of garlic juice of 0.5 ml/200gBB garlic juice with 25% garlic juice concentration is an effective volume in reducing protease enzyme activity by 43% and preventing damage to histopathologic images of rat’s jejunum (Rattus norvegicus).
