Abstract

Hiperkolesterolemia merupakan kondisi dimana terjadi peningkatan kolesterol dalam darah, yang nantinya akan memicu peningkatan radikal bebas dan menyebabkan kerusakan endotel ginjal sehingga mengalami albuminuria. Tomat diketahui memiliki kandungan likopen yang berpotensi sebagai antioksidan. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh ekstrak buah tomat untuk meningkatkan kadar albumin dan menekan kerusakan pada ginjal tikus (Rattus norvegicus) model Hiperkolesterolemia umur 8-12 minggu dengan bobot 150-200 gram yang dibagi menjadi 5 kelompok perlakuan yang diberikan diet hiperkolesterolemia selama 14 hari dan dilanjutkan dengan terapi selama 14 hari dengan dosis 200 , 400, dan 600 mg/Kg BB. Variabel yang diamati adalah kadar albumin darah dan gambaran histopatologi ginjal. Adapun kadar albumin dianalisa secara kualitatif dengan ragam analisa ANOVA yang dilanjutkan dengan Uji Tukey (α=0,05) menggunakan pengujian Bromcresol green dan gambaran histopatologi ginjal diamati dengan menggunakan pewarnaan HE untuk melihat kerusakan yang terjadi pada ginjal. Hasil penelitian menunjukkan bahwa terapi ekstrak buah tomat dapat meningkatkan kadar Albumin yaitu dari 1.45±0.006 pada kondisi hiperkolestrolemia menjadi 2.47±0.004 pada terapi dosis 600 mg/kg BB. Gambaran histopatologi ginjal menunjukkan adanya perbaikan pada glomerulus jika dibandingkan dengan tikus hiperkolestrolemia. Dari penelitian ini dapat disimpulkan bahwa terapi ekstrak buah tomat dengan dosis 600 mg/kg BB dapat meningkatkan kadar Albumin dan memperbaiki kerusakan ginjal tikus hiperkolestrolemia. Ekstrak air buah tomat dengan dosis 600 mg/kg BB menunjukkan peningkatan kadar Albumin pada tikus hiperkolestrolemia tertinggi yaitu 23,79%.

English Abstract

Hypercholesterolemia is a condition of an increase of cholesterol inside the blood, which will lead to an increase of free radicals and causing endothelial damage on kidneys wich will trigger albuminuria. Tomato is known to have lycopene which has potential as antioxidant. The purpose of this study was to determine the effect of tomato extracts to increase the levels of albumin and reduce the damage of the kidneys of rats (Rattus norvegicus) model of hypercholesterolemia. This study used 20 strain wistar rats aged between 10 – 12 weeks with the weight around 150 – 200 grams which were divided into negative control group,positive control group(hypercholesterolemia) as well as group which given the tomato extract therapy doses at 200, 400 and 600 mg/kg BB. Tomato extract is given for 14 days after the distribution of hypercholesterol diet. The variables measured were the albumin levels inside the blood and renal histophatology picture. The albumin level quantitatively analyzed with the type of ANOVA analysis followed by Tukey test (α=0,05) using Bromocresol green test and renal histophatology picture observed using HE staining to see the damage the occurs on the kidneys. The results showed that treatment of tomato fruit extract can increase levels of Albumin are of 1.45±0.006 on the condition hiperkolestrolemia be 2.47±0.004 in therapeutic dose of 600 mg / kg. Histopathologic features kidneys showed a reduce damage of glomerulus compared to mice hiperkolestrolemia. From this study it can be concluded that the therapy tomato fruit extract can increase levels of Albumin and repair damage hiperkolestrolemia rat kidneys. Water extract of tomatoes at a dose of 600 mg / kg body weight more effectively increase levels of Albumin in rats hiperkolestrolemia the highest was 23,79%.
