Abstract

Latar belakang: Sklerostin adalah protein pada manusia yang terbukti mempercepat apoptosis osteoblas sehingga menurunkan kemampuan pembentukan tulang yang mengakibatkan terjadinya osteoporosis. Sel dendritik adalah monosit yang terdiferensiasi oleh stimulasi GM-CSF dan IL-4, dan menjadi bagian sistem kekebalan mamalia. Pada keadaan osteoporosis jumlah sel dendritik meningkat. Tujuan penelitian ini adalah mengetahui efek sklerostin terhadap jumlah sel dendritik pada limpa tikus putih (Rattus norvegicus) galur Wistar model hipoestrogen. Metode: Penelitian ini menggunakan desain (true experimental) randomized post test only controlled group menggunakan 24 ekor tikus. Tikus dibagi 6 kelompok, yaitu: kontrol negatif, kontrol positif, kelompok perlakuan diberi injeksi protein sklerostin dengan dosis 0.1 μg, 1 μg, 2 μg, dan 100 μL ajuvan Freund’s saja. Perhitungan jumlah sel dendritik dilakukan dengan metode immunohistokimia. Hasil: Terdapat penurunan jumlah sel dendritik yang bermakna pada semua kelompok perlakuan kecuali Kelompok KN dengan P3 tidak menunjukkan perbedaan yang bermakna dengan nilai (p = 0,052 ; α < 0,05). Pada penelitian ini didapatkan hasil korelasi negatif yang bermakna antara pemberian protein sklerostin beserta ajuvan Freund’s dengan jumlah sel dendritik (p = 0,003, r = 0,694). Kesimpulan: Mengetahui efek pemberian protein sklerostin terhadap jumlah sel dendritik. Pemberian protein sklerostin dan ajuvan Freund’s dapat menurunkan jumlah sel dendritik pada tikus putih (Rattus norvegicus) galur Wistar model hipoestrogen.

English Abstract

Background: Sclerostin is a protein in humans that proves to accelerate apoptosis of osteoblasts, thereby decreasing the ability of bone formation, resulting in osteoporosis. Dendritic cells are monocytes differentiated by GM-CSF and IL-4 stimulation, and are part of the mammalian immune system. In osteoporosis the number of dendritic cells increases. To find out study aims to is that sclerostin can decrease the number of dendritic cells in the white rat spleen (Rattus norvegicus) Wistar strain of the hypoestrogen model. Method: This study used a true experimental randomized post test only in the controlled group design using 24 rats. Rats were divided into 6 groups, namely: negative control, positive control, treatment group given injection of sclerostin protein with dose of 0.1 μg, 1 μg, 2 μg, and 100 μL adjuvant Freund's alone. Calculation of the number of dendritic cells was performed by immunohistochemical methods. Results: There was a significant decrease in the number of dendritic cells in all treatment groups except groups KN with P3 that did not show significant differences with values (p = 0.052; α <0.05). In this study, there was a significant negative correlation between sclerostin protein and Freund's adjuvant with dendritic cell count (p = 0,003, r = 0,694). Conclusion: Determines the effect of protein sclerostin on the number of dendritic cells. Administration of Freund's protein sclerostin and adjuvant may decrease the number of dendritic cells in white rats (Rattus norvegicus) Wistar strains of the hypoestrogen model.
