Abstract

The Myostatin (MSTN) gene, a member of the transforming growth factor-β (TGF-β) superfamily, acts as a negative regulator of muscle mass. This research aimed to identify polymorphisms in the MSTN gene in Ongole Crossbred (PO) and Madura cattle population using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCRRFLP) analysis. A total of 122 DNA samples were analyzed, consisting of 64 PO cattle from UPTPT HMT Tuban and 58 Madura cattle from Pamekasan, Madura Island. DNA restriction fragment length polymorphism (RFLP) targeted SNPc.267G>A along a 541 bp DNA sequence amplified using PCR. The forward primer 5’- CAAGTTGTCTCTCAGACTGG-3’ and reverse primer 5’- CTAGAACAGCAGTCAGCAGA-3’ were used with anannealing temperature of 61°C. The ALU1 restriction enzyme successfully digested the target sequence, producing three polymorphic genotypes: AA, GG, and AG. The results showed allele frequencies in Ongole Crossbred (PO) cattle were 0.422 (G) and 0.578 (A), with genotype frequencies of 0.234 (GG), 0.375 (AG), and 0.391 (AA). Whereas, Madura cattle exhibited allele frequencies of 0.216 (G) and 0.784 (A), with genotype frequencies of 0.069 (GG), 0.293 (AG), and 0.638 (AA). Observed heterozygosity (Ho) was lower than expected heterozygosity (He) in both breeds, with values of 0.375 (Ho) and 0.488 (He) for PO, and 0.293 (Ho) and 0.338 (He) for Madura. These findings highlight significant variations in allele and genotype distributions between the two breeds, with both populations in Hardy-Weinberg, and show moderate informative polymorphism information content value.
