Abstract
Untuk mengetahui efek kombinasi ekstrak kopi hijau dan ekstrak teh hijau (CM) terhadap kekeruhan lensa, ekspresi, dan agregasi lensa αB-crystallin pada model tikus Sindrom Metabolik (SM). METODE Penelitian eksperimental melibatkan dua puluh empat ekor tikus Sprague Dawley yang dibagi menjadi empat kelompok (kontrol positif, kontrol negatif, kombinasi terdekafeinasi (dekaf) dan tanpa dekafeinasi (non-dekaf). Setelah berkembang menjadi SM, kelompok perlakuan diberikan ekstrak teh hijau (GT) 0,3 gram/kgBB + ekstrak kopi hijau (GC) 0,2 gram/kgBB terdekafeinasi (n=6) dan GT 0,3 gram/kgBB + GC 0,2 gram/kgBB non-dekaf (n=6) selama sembilan minggu. Derajat kekeruhan lensa diperiksa dengan mikroskop binokular, ekspresi serta agregasi αBcrystallin menggunakan western blot. Kruskal Wallis, one way ANOVA, dan korelasi dan regresi digunakan. HASIL Tidak terdapat perbedaan rerata derajat katarak (p=0,202>0,050) maupun korelasi (p 0,302>0,050) antara kelompok perlakuan dan kontrol. Kelompok dekaf meningkatkan ekspresi αBcrystallin native secara signifikan (p=0,00<0,05) sebesar 86,62% dengan R2 sebesar 87,2%, sedangkan dekaf maupun non-dekaf tidak mempengaruhi agregasi (p 0,054>0,050). KESIMPULAN Baik CM dekaf maupun non-dekaf tidak memengaruhi tingkat kekeruhan lensa. Namun, kombinasi dekaf lebih meningkatkan lensa asli αB-kristalin dibandingkan kelompok non-dekaf. Meskipun demikian, kombinasi kedua ekstrak tidak berdampak pada agregasi αB-Crystalline.
English Abstract
To investigate the potential effect of combination green coffee and green tea extract (CM) on the lens opacity, expression, and the aggregation of lens αBcrystallin in a rat model of Metabolic Syndrome (MetS). METHODS This is an experimental study on twenty-four Sprague Dawley rats which divided into four groups (positive control, negative control, a combination which was decaffeinated (decaf); and non-decaffeinated (non-decaf). After developing MetS, the treatment groups were administered a green tea extract (GT) of 0.3 gram/kgBB + green coffee extract (GC) of 0.2 gram/kgBB decaf (n=6) and an GT of 0.3 gram/kgBB + GC 0.2 gram/kgBB non-decaf (n=6) for nine weeks. The degree of lens opacity was examined with a binocular microscope, and lens αBcrystallin expression and aggregation were measured from western blot analysis. Kruskal Wallis, one-way ANOVA and regression statistical analysis were used. RESULTS There were no differences in mean cataract gradings (p=0.202>0.050) nor correlation (p 0.302>0.050) between the treatment and control groups. The decaf promotes the expression of native lens αB-crystallin, with a significant difference between the control and non decaf group (p=0.00<0.05). Also, it enhances expression by 86.62% with an R2 of 87.2%, whereas the combination which decaf and non decaf did not affect the aggregation (p 0.054>0.050). CONCLUSION Neither decaffeinated nor caffeinated CM affected lens opacification grade. However, the decaffeinated combination promotes native lens αB-crystallin more than the caffeinated combination. Nevertheless, the combination of both extracts had no impact on αB-crystallin aggregation.
