Abstract

TUJUAN Untuk mengetahui pengaruh kombinasi ekstrak teh hijau dan ekstrak kopi hijau pada kadar tumor necrosis factor alpha (TNFa) di vitreous pada tikus model sindroma metabolik. METODE Penelitian eksperimental melibatkan dua puluh empat ekor tikus Sprague Dawley yang dibagi menjadi empat kelompok (kontrol positif, kontrol negatif, kombinasi terdekafeinasi (dekaf) dan tanpa dekafeinasi (non-dekaf). Setelah berkembang menjadi SM, kelompok perlakuan diberikan ekstrak teh hijau (GT) 0,3 gram/kgBB + ekstrak kopi hijau (GC) 0,2 gram/kgBB terdekafeinasi (n=6) dan GT 0,3 gram/kgBB + GC 0,2 gram/kgBB non-dekaf (n=6) selama sembilan minggu. Kadar TNF-a di vitreous diperiksa menggunakan metode ELISA. Data kemudian dioleh menggunakan one-way ANOVA dan post hoc multiple comparison Tukey. HASIL Didapatkan penurunan kadar TNF-a di vitreous pada kelompok perlakuan terdekafeinasi (P=0.415>0.050) dan perlakuan tanpa dekafeinasi (P=0.415>0.196) meskipun tidak berbeda signifkan dengan kelompok kontrol positif. Kelompok perlakukan terdekafeinasi memberikan hasil lebih baik daripada kelompok perlakuan tanpa dekafeinasi meskipun tidak berbeda signifikan (P=0.961>0.050) . KESIMPULAN Kombinasi ekstrak teh hijau dan ekstrak kopi hijau terdekafeinasi maupun tanpa dekafeinasi dapat menurunkan kadar tumor necrosis factor alpha (TNFa) di vitreous pada tikus model sindroma metabolik meskipun masih tidak berbeda signifikan dengan kontrol positif.

English Abstract

OBJECTIVES To investigate the potential effect of combination green coffee and green tea extract (CM) on Tumor Necrosis Factor Alpha Levels in the Vitreous in a rat model of Metabolic Syndrome (MetS). METHODS This is an experimental study on twenty-four Sprague Dawley rats which divided into four groups (positive control, negative control, a combination which was decaffeinated (decaf); and non-decaffeinated (non-decaf). After developing MetS, the treatment groups were administered a green tea extract (GT) of 0.3 gram/kgBB + green coffee extract (GC) of 0.2 gram/kgBB decaf (n=6) and an GT of 0.3 gram/kgBB + GC 0.2 gram/kgBB non-decaf (n=6) for nine weeks. TNF-a levels in the vitreous were examined using the ELISA method. The data then processed using one-way ANOVA and Tukey's post hoc multiple comparison. RESULTS There was a decrease in TNF-a levels in the vitreous in the decaffeinated treatment group (P=0.415>0.050) and non-decaffeinated treatment (P=0.415>0.196) although it was not significantly different from the positive control group. The decaffeinated treatment group gave better results than the non-decaffeinated treatment group, although it was not significantly different (P=0.961>0.050). CONCLUSION The combination of green tea extract and decaffeinated or non-decaffeinated green coffee extract can reduce levels of tumor necrosis factor alpha (TNFa) in the vitreous in metabolic syndrome model mice, although it is still not significantly different from the positive control.
