Abstract
Latar Belakang: Karsinoma Nasofaring (KNF) adalah tumor ganas yang berasal dari epitel mukosa nasofaring di fossa Rossenmuller. KNF merupakan penyakit yang endemik di Cina Selatan, Asia Tenggara dan Afrika Utara. Ekspresi seluler cyclooxygenase-2 (COX-2) dan vascular endothelial growth factor (VEGF) ditemukan meningkat pada proses limfangiogenesis sel kanker yang diukur melalui pembesaran kelenjar getah bening leher. Penilaian COX-2 dan VEGF pada tumor memerlukan penerapan indikator pemeriksaan MVD yang dapat diukur dengan menggunakan antigen spesifik yang ada pada endotel vaskular yaitu CD105. Ekspresi seluler COX-2 ditemukan berkorelasi positif dengan ekspresi VEGF pada penderita karsinoma nasofaring. Tujuan: Menganalisis adanya korelasi antara ekspresi COX-2 dengan VEGF pada karsinomanasofaring tipe undifferentiated. Metode: Penelitian ini adalah penelitian observasional analitik dengan desain cross sectional yang melibatkan 24 sampel biopsi jaringan nasofaring dari penderita KNF tipe undifferentiated. Pemeriksaan ekspresi COX-2 dan VEGF menggunakan marker MVD (CD-105) yang menggunakan pewarnaan imunohistokimia yang dinilai oleh ahli Patologi Anatomi. Sedangkan Kelenjar getah bening leher diukur menggunakan rumus ellipsoid formula; yakni {volume=(π*(x*y*z)/6)}. Hasil: Hasil uji korelasi COX-2 dan VEGF tidak memiliki hubungan signifikan dengan nilai r = -0.074, p = 0.732 (>0,05). COX-2 dengan pembesaran kelenjar getah bening tidak memiliki hubungan signifikan dengan nilai korelasi -0,052 dan P=0,808. VEGF dengan pembesaran kelenjar getah bening tidak memiliki hubungan signifikan korelasi -0,011 dan P=0,958. Kesimpulan: Tidak didapatkan korelasi ekspresi COX-2 dan VEGF pada karsinoma nasofaring tipe undifferentiated. Tidak didapatkan korelasi ekspresi COX-2 terhadap pembesaran kelenjar getah bening leher. Tidak didapatkan korelasi ekspresi VEGF terhadap pembesaran kelenjar getah bening leher.
English Abstract
Background: Nasopharyngeal Carcinoma (NPC) is a malignant tumor originating from the mucosal epithelium of the nasopharynx in the Rossenmuller fossa. NPC is a disease that is endemic in South China, Southeast Asia and North Africa. Cellular expression of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) was found to increase in the lymphangiogenesis process of cancer cells as measured by enlargement of cervical lymph nodes. Assessment of COX- 2 and VEGF in tumors requires the application of MVD examination indicators which can be measured using a specific antigen present on the vascular endothelium, namely CD105. COX-2 cellular expression was found to be positively correlated with VEGF expression in nasopharyngeal carcinoma patients. Objective: To analyze the correlation between COX-2 and VEGF expression in undifferentiated type of nasopharyngeal carcinoma. Method: This study was an analytical observational study with a cross sectional design involving 24 nasopharyngeal tissue biopsy samples from patients with undifferentiated type of NPC. COX-2 and VEGF expression examination uses the MVD marker (CD-105) using immunohistochemical staining assessed by an anatomical pathologist. Meanwhile, cervical lymph nodes are measured using the ellipsoid formula; namely {volume=(π*(x*y*z)/6)}. Results: The results of the COX-2 and VEGF correlation test did not have a significant relationship with a value of r = -0.074, p = 0.732 (>0.05). COX-2 with enlarged lymph nodes does not have a significant relationship with a correlation value of -0.052 and P=0.808. VEGF and lymph node enlargement did not have a significant correlation -0.011 and P=0.958. Conclusion: There was no correlation between COX-2 and VEGF expression in undifferentiated type of nasopharyngeal carcinoma. There was no correlation between COX-2 expression and enlargement of cervical lymph nodes. There was no correlation between VEGF expression and enlargement of cervical lymph nodes.
