Abstract
TUJUAN Untuk mengetahui pengaruh pemberian kombinasi ekstrak teh hijau dan ekstrak kopi hijau terhadap panjang aksial bola mata terhadap biomekanik sklera dan ekspresi NF-κB sklera pada tikus sindroma metabolik METODE Penelitian eksperimental melibatkan dua puluh empat ekor tikus Sprague Dawley yang dibagi menjadi empat kelompok (kontrol positif, kontrol negatif, kombinasi terdekafeinasi (dekaf) dan tanpa dekafeinasi (non-dekaf). Setelah berkembang menjadi SM, kelompok perlakuan diberikan ekstrak teh hijau (GT) 0,3 gram/kgBB + ekstrak kopi hijau (GC) 0,2 gram/kgBB terdekafeinasi (n=6) dan GT 0,3 gram/kgBB + GC 0,2 gram/kgBB non-dekaf (n=6) selama sembilan minggu. Biomekanik sklera diukur dengan panjang aksial bola mata dan ekspresi NF-κB diukur dengan pewarnaan menggunakan imunohistokimia dengan reagen NF-κB p65 (7G6) Monoclonal Antibody . Data kemudian diolah dengan uji komparatif dan korelasi antar kelompok. HASIL Didapatkan penurunan ekspresi NF-κB yang signifikan pada kelompok dekaf dan non dekafeinasi dibandingkan kelompok kontrol positif (p=0.00,p<0.05). Selain itu, didapatkan penurunan panjang aksial bola mata pada kelompok dekaf dan non dekaf namun tidak signifikan (p=0.06 dan p=0.07,p>0.05). Sebagai luaran sekunder didapatkan tebal sklera tidak berbeda signifikan antara kontrol positif, kelompok dekaf dan non dekaf. KESIMPULAN Pemberian kombinasi ekstrak teh hijau dan ekstrak kopi hijau baik terdekafeinasi dan tanpa dekafeinasi dapat secara signifikan menurunkan ekspresi NF-κB sklera pada tikus model sindroma metabolik.
English Abstract
PURPOSE To determine the effect of the combination of green tea extract and green coffee extract on the axial length of the eyeball on sclera biomechanics and sclera NF-κB expression in rats model of metabolic syndrome (Mets) METHOD The experimental study involved twenty-four Sprague Dawley rats divided into four groups (positive control, negative control, decaffeinated (decaf) and decaffeinate (non-decaffeinated) combinations. After developing to Mets, the treatment group was given green tea extract (GT) 0.3 grams/kgBB + green coffee extract (GC) 0.2 grams/kgBB decaffeinated (n = 6) and GT 0.3 grams/kgBB + GC 0.2 grams/kgBB non-decaffeinated (n = 6) for nine weeks. The biomechanics of the sclera were measured by the axial length of the eyeball and the expression of NF-κB was measured by staining using immunohistochemistry with the reagent NF-κB p65 (7G6) Monoclonal Antibody. The data processed with comparative tests and correlations test between groups. RESULT There was a significanty decrease in NF-κB expression in the decaf and nondecaffeination groups compared to the positive control group (p = 0.00, p < 0.05). In addition, there was a decrease in the axial length of the eyeball in the decaf and non-decaf groups when compared to the positive group but not significant (p = 0.06 and p = 0.07, p > 0.05). As a secondary output, sclera thickness was not significantly different between positive controls, decaf and non-decaf groups. CONCLUSION The combination of green tea extract and green coffee extract, decaffeinated and decaffeinate can significantly decrease the expression of sclera NF-κB in rat models of metabolic syndrome.
