Abstract
PCOS (Polycystic ovary syndrome) merupakan masalah reproduksi yang dialami oleh wanita usia reproduktif. Manifestasi klinis penderita PCOS sangat bervariasi karena gangguan PCOS seringkali diikuti dengan kelainan metabolik dan endokrin. Terganggunya metabolisme hepar yang disebabkan tingginya kadar estrogen akibat PCOS terlihat adanya radikal bebas yang dideteksi dengan kadar Malondialdehid (MDA) hepar dan perubahan histopatologis hepar tikus. Penelitian ini bertujuan untuk mengetahui pengaruh testosteron propionat dan estradiol valerat terhadap kadar MDA hepar dan perubahan histopatologis hepar tikus. Penelitian ini dilakukan pada bulan Juni hingga Oktober 2022. Penelitian bersifat eksperimental menggunakan tikus putih betina galur Wistar berumur 6 bulan dengan berat badan ratarata 130-180 gram. Hewan coba dibagi menjadi tiga kelompok perlakuan yaitu kontrol negatif yang diinduksi NaCl fisiologis, kelompok perlakuan 1 yang diinduksi testosteron propionat dosis 100 mg/KgBB selama 12 hari secara intraperitoneal, dan kelompok perlakuan 2 yang diinduksi estradiol valerat dosis 2 mg/KgBB selama 2 hari. Data dianalisis secara kualitatif dan hasil pengamatan histopatologis akan dianalisis menggunakan bantuan software komputer yaitu ImageJ. Data kualitatif dilakukan dengan uji One Way ANOVA dan dilanjutkan dengan Uji Jarak Berganda Duncan. Hasil analisis data menunjukkan rata-rata MDA hepar yang diinduksi testosteron propionat (99,47 ng/ml), estradiol valerat (50,80 ng/ml) dan kontrol (69,33 ng/ml). Uji One Way ANOVA menyatakan p > 0,05 yang berarti tidak terdapat perbedaan yang nyata. Namun pada Uji Jarak Berganda Duncan terlihat perbedaan yang nyata pada perlakuan 1 terhadap perlakuan 2 namun tidak tampak perbedaan yang nyata pada kelompok kontrol terhadap perlakuan 1 maupun kelompok kontrol terhadap perlakuan 2. Ratarata skoring degenerasi melemak hepar yang diinduksi testosteron propionat (2) dan estradiol valerat (1,83) mengalami kenaikan daripada keadaan normal (1,33). Uji Kruskal Walis menunjukkan p ≤ 0,05 sehingga dinyatakan berbeda signifikan terhadap perlakuan kontrol.
English Abstract
PCOS (Polycystic ovary syndrome) is a reproductive problem experienced by women of reproductive age. The clinical manifestations of PCOS sufferers vary greatly because PCOS disorders are often accompanied by metabolic and endocrine disorders. Disruption of hepatic metabolism caused by high levels of estrogen due to PCOS can be seen by the presence of free radicals which are detected by hepatic malondialdehyde (MDA) levels and histopathological changes in rat livers. This study aims to determine the effect of testosterone propionate and estradiol valerate on hepatic MDA levels and histopathological changes in rat liver. This research was conducted from June to October 2022. This research was experimental using Wistar strain female white rats aged 6 months with an average body weight of 130-180 grams. Experimental animals were divided into three treatment groups: negative control induced by physiological NaCl, treatment group 1 which was induced intraperitoneally by testosterone propionate dose of 100 mg/KgBB for 12 days, and treatment group 2 which was induced by estradiol valerate dose of 2 mg/KgBB for 2 days. Data were analyzed qualitatively and the results of histopathological observations will be analyzed using the help of computer software, namely ImageJ. Qualitative data is carried out by the One Way ANOVA test and continued with Multiple Range Test (Duncan test). The results of the data analysis show that the average hepatic MDA induced by testosterone propionate (99,47 ng/ml), estradiol valerate (50,80 ng/ml) and control (69,33 ng/ml). The One Way ANOVA test states p > 0,05, which means there is no significant difference. However, in Duncan’s Multiple Range Test, there was a significant difference between treatment 1 and treatment 2, but there was no significant difference in the control group compared to treatment 1 or the control group compared to treatment 2. The average scoring of hepatic fatty degeneration induced by testosterone propionate (2) and estradiol valerate (1,83) increased compared to normal (1,33). The Kruskal Walis test showed p ≤ 0,05 so it was declared significantly different from the control treatment
