Abstract
Tujuan dari penelitian yang dilakukan adalah memperoleh presentase penggunaan gum arab (Acacia senegal) terbaik sebagai mikroenkapsulan susu bubuk kuda Sumbawa dan mengetahui potensi peptida bioaktifnya secara analisis in silico sebagai functional healthy food. Metode peneltian yang dilakukan menggunakan rancangan acak lengkap (RAL) dengan perhitungan Analysis of Variance (ANOVA) yang terdiri dari 4 perlakuan dan 4 ulangan. Hasil rataan data yang berbeda nyata atau sangat nyata akan dilanjutkan dengan analisis Uji Jarak Berganda Duncan (UJBD). Perlakuan penggunaan gum arab yang dilakukan adalah P1 (20%), P2 (24%), P3 (28%) dan P4 (32%). Variabel yang dianalisis terdiri dari total BAL, TPC, pH, total asam laktat tertitrasi, antibakteri, antioksidan, jumlah E. coli, mikrostruktur, gugus fungsi, kandungan protein dan in silico. Penggunaan gum arab sebagai mikroenkapsulan susu bubuk kuda Sumbawa memberikan pengaruh yang sangat nyata (P<0,01) terhadap pH, total asam laktat tertitrasi, aktivitas antibakteri (E. coli, S. thypimurium, dan S. aureus) dan aktivitas antioksidan. Hasil analisis total BAL dan TPC memberikan pengaruh yang nyata (P<0,05), sedangkan aktivitas antibakteri pada bakteri uji L. casei memberikan pengaruh yang tidak nyata (P>0,05). Hasil analisis dari jumlah E. coli, mikrostruktur partikel FESEM/EDX, uji gugus fungsi protein dengan FTIR, kandungan protein dengan LCMS/MS dan in silico menggunakan metode deskriptif. Hasil penelitian analisis in silico, didapatkan rantai pendek peptida bioaktif RP, SF, HP dan RY sebagai calon ligan DPP–IV Inhibitor ACE–Inhibitor dan Antioksidan pada k– Kasein susu bubuk kuda Sumbawa. Hasil akhir pada penelitian yang telah dilakukan bahwa uji in silico dapat menyimulasikan komposisi senyawa bioaktif susu bubuk kuda Sumbawa terutama k–Kasein, untuk memprediksi potensi functional healthy food
English Abstract
The purpose of this study was to obtain the best percentage of gum arabic (Acacia senegal) as a microencapsulant of Sumbawa horse milk powder and to determine the potential of its bioactive peptides by in silico analysis as a functional healthy food. The research method used a complete randomised design (CRD) with Analysis of Variance (ANOVA) calculation consisting of 4 treatments and 4 replicates. The results of data averages that are significantly or very significantly different will be continued with the analysis of Duncan's Multiple Range Test (DMRT). The treatments using gum arabic were T1 (20%), T2 (24%), T3 (28%) and T4 (32%). The variables analysed consisted of total LAB, TPC, pH, total lactic acid titratable, antibacterial, antioxidant, E. coli count, microstructure, functional groups, protein content by LCMS/MS and in silico. The use of gum arabic as microencapsulant of Sumbawa horse milk powder gave a very significant effect (P<0.01) on pH, total lactic acid titratable, antibacterial (E. coli, S. thypimurium, and S. aureus) and antioxidant. The analysis results of total LAB and TPC gave a significant effect (P<0.05), while L. casei antibacterial gave no significant effect (P>0.05). The results were analysed from the number of E. coli, particle microstructure FESEM/EDX, protein functional group test with FTIR, protein content with LCMS/MS and in silico using descriptive methods. The results of in silico analysis research, obtained short chains of bioactive peptides RP, SF, HP and RY as prospective ligands of DPP–IV Inhibitor ACE– Inhibitor and Antioxidant in k–Casein of Sumbawa horse milk powder. The final result of the research that has been carried out is that the in silico test can simulate the composition of bioactive compounds of Sumbawa horse milk powder, especially k–Casein, to predict the potential of functional healthy food
