Abstract

Protein TPO atau Thyroid Peroxidase berperan penting Dalam menjalankan fungsi fisiologis pada kelenjar tiroid defesiensi protein ini dapat menyebabkan kecacatan fungsi kelenjar tiroid yang akan menyebabkan kelenjar tiroid tidak dapat menghaslkan hormon tiroid. Kekurangan hormon tiroid dapat menyebabkan gangguan pada proses metabolisme sehingga dapat mengganggu tumbuh kembang manusia. Penelitian ini bertujuan untuk melalukan isolasi protein TPO 52 kDa melalui metode SDS-PAGE dan menguji imunogenesitas TPO 52 kDa melalui reaksi positif dengan antibodi hasil induksinya dengan metode Dot Blot. Penelitian bersifat eksperimental dengan menggunakan hewan coba kelinci dari galur New Zealand White yang berkelamin jantan, berat 3 kg, dan berumur 6 bulan. Kelinci dikelompokan menjadi 2 kelompok, yaitu kelinci kontrol dan kelinci yang diinduksi protein TPO 52 kDa.

English Abstract

The TPO protein, or Thyroid Peroxidase, plays a crucial role in the physiological functions of the thyroid gland. A deficiency in this protein can lead to impaired thyroid gland function, resulting in the gland's inability to produce thyroid hormones. A lack of thyroid hormones can disrupt metabolic processes, thereby affecting human growth and development. This study aims to investigate the immunogenicity of the 52 kDa TPO protein using SDS-PAGE and dot blot methods with anti-TPO antibody after induction. The experimental research utilized male New Zealand White rabbit, weighing 3 kg and aged 6 month. The rabbit were divided into two groups: a control group and a group induced with TPO protein. The 52 kDa TPO protein induction performed three times.
