Abstract

Latar Belakang: Staphylococcus aureus merupakan jenis organisme gram positif penyebab terjadinya infeksius hingga abses bernanah yang berakibat jaringan akan mengalami nekrosis. Bakteri patogen seperti Staphylococcus aureus sering kali tahan pada beberapa antibiotik. Oleh karena itu, alternatif tradisional dengan menggunakan tanaman seledri, dapat digunakan sebagai pendamping antibiotik. Seledri memiliki kemampuan membunuh bakteri dengan kandungan, seperti flavonoid, saponin, dan tanin. Tujuan: tujuan dari studi ini adalah menguji sifat antibakteria dari ekstrak etanol seledri (Apium graveolens L) terhadap Staphylococcus aureus dengan cara in vitro. Metode: Difusi sumuran digunakan untuk melihat zona hambat Staphylococcus aureus. Hasil dan Pembahasan: Dari pengukuran zona yang menghambat pertumbuhan Staphylococcus aureus menghasilkan berbagai konsentrasi ekstrak seledri yaitu 50%, 60%, 70%, 80%, 90%, dan 100% yang memiliki hambatan bening dengan rata-rata 6mm; 7mm; 7.5mm; 8.5mm; 11.25mm, dan 12mm. Namun, hasil data tersebut lebih rendah daripada Ampicillin 1g yang menunjukan rata rata zona hambat 34.25mm. Data inidianalisis menggunakan uji non parametrik Kruskal- Wallis. Hasil Kruskal-Wallis ada perbedaan yang bermakna pada hambatan Staphylococcus aureus pada tiap konsentrasi p-value = 0.001. Sehingga dapat diteruskan menggunakan uji Mann Whitney untuk menentukan perbedaan signifikan antar perlakuan. Studi ini juga menggunakan uji Spearman yang menunjukkan R=0.886 (R>0.5) konsentrasi ekstrak etanol seledri dengan zona hambat Staphylococcus aureus memiliki hubungan yang sangat kuat. Kesimpulan: Kesimpulan pada studi ini yaitu ekstrak etanol seledri (Apium graveolens L) terbukti menunjukkan aktivitas antibakteri terhadap pertumbuhan Staphylococcus aureus secara in vitro

English Abstract

Background: Staphylococcus aureus is a gram-positive bacteria causing infections and purulent abscesses and that makes tissues necrosis. Pathogenic bacteria like Staphylococcus aureus often increasing antibiotics resistance. So, using a traditional medicine as an alternative like Celery can be used. Celery contains ingredients that potentially can inhibit bacterial growth, namely flavonoids, saponins and tannins. Objective: The aim of this study was to test the antibacterial effect of celery (Apium graveolens L) ethanol extract against Staphylococcus aureus in vitro. Method: In this study, well diffusion was used to determine the ihnhibition zone of Staphylococcus aureus. Results and Discussion: Inhibition zone of celery extract to the Staphylococcus aureus growth showed on various concentrations 50%, 60%, 70%, 80%, 90%, and 100% with an average inhibition zone 6mm ; 7mm; 7.5mm; 8.5mm; 11.25mm, and 12mm. All data results were smaller than the positive control, Ampicillin 1g with an average inhibition zone 34.25mm. This data were analyzed using the Kruskal- Wallis statistical test. There was a significant difference in the inhibition zone of Staphylococcus aureus at each concentration (p-value = 0.001). So it can be continued with the Mann Whitney test to determine significant differences between treatments. Moreover, this research used the Spearman test with a value of R = 0.886 (R > 0.5) indicating that there is a strong correlation between the concentration of celery ethanol extract and the inhibition zone of Staphylococcus aureus. Conclusion: This study proves that celery ethanol extract (Apium graveolens L) has been shown to have an antibacterial effect on the of Staphylococcus aureus by in vitro method
