Abstract

Temulawak (Curcuma xanthorrizha Roxb) merupakan tumbuhan rhizoma khas Indonesia yang bermanfaat menangkal radikal bebas karena memiliki senyawa antioksidan seperti senyawa fenolik dan flavonoid. Ekstraksi dilakukan menggunakan metode microwave assisted extraction (MAE) dengan pelarut etanol 70% sebagai metode dan pelarut terbaik untuk mengekstrak fenol dan flavonoid yang diperoleh dari tahap preliminary. Tujuan penelitian ini adalah untuk mengetahui rasio pelarut, daya ekstraksi, dan waktu ektraksi optimal menggunakan Response Surface Methodology. Rancangan penelitian yang digunakan adalah Response Surface Methodology (RSM) rancangan Box-Behnken Design (BBD) dengan tiga variabel bebas yaitu rasio pelarut, daya ekstraksi, dan waktu ekstraksi. Respon yang dianalisis yaitu total fenol dan total flavonoid. Titik minimum dan maksimum rasio pelarut, daya ekstraksi, dan waktu ekstraksi diatur sebesar 30 – 90 mL/gram, 150 – 450 watt, dan 1 – 5 menit secara berturut-turut. Kondisi operasi optimal yang diperoleh dari software DesignExpert12 kemudian divalidasi. Pengujian yang dilakukan pada penelitian ini meliputi uji total fenol dan uji total flavonoid. Hasil menunjukkan bahwa rasio pelarut memberikan pengaruh signifikan terhadap total fenol dan daya ekstraksi memberikan pengaruh signifikan teradap total flavonoid. Proses optimum ekstraksi berada pada kondisi operasi rasio pelarut 66mL/gr, daya ekstraksi 450 watt, dan waktu ekstraksi 1 menit dengan karakteristik ekstrak yaitu total fenol 21.4868±3.6169 mg GAE/g dw dan total flavonoid 26.0382±5.69773 mg QE/g dw. Hasil verifikasi menunjukkan total fenol 12.93451±1.425 mg GAE/g dw dan total flavonoid 21.79954±0.9847 mg QE/g dw. Hasil verifikasi tidak sesuai dengan prediksi yang ditunjukkan dengan nilai p-value kurang dari 0.05 menunjukkan bawa model yang tidak akurat memprediksi hasil aktual.

English Abstract

Temulawak (Curcuma xanthorrizha Roxb) is a typical Indonesian rhizome plant which is useful for warding off free radicals because it has antioxidant compounds such as phenolic and flavonoids compounds. Temulawak was extracted using microwave assisted extraction (MAE) method with ethanol 70% solvent as the best method and solvent extraction to extract the phenolic and flavonoid content resulted from preliminary. The objective of this study is to analyze solvent ratio, extraction power, and extraction time using Response Surface Methodology (RSM). Experimental design used in this study is Response Surface Methodology (RSM) Box-Behnken Design (BBD) with three independent variables such as solvent ratio, extraction power, and extraction time. Responses that were analyzed including total phenol and total flavonoid. The minimum and maximum point of solvent ratio, extraction power, and extraction time were set at 30 – 90 mL/gram, 150 – 450 watt, and 1 – 5 minutes, consecutively. The optimum operation condition obtained from DesignExpert12 were then validated. The tests that were used in this study including total phenolic content test and total flavonoid content test. Result showed that solvent ratio has a significant effect towards total phenol and extraction power has a significant effect towards total flavonoid. The optimum extraction processes are under operating condition with solvent ratio of 66mL/g, extraction power of 450 watt, and extraction time of 1 minute with the characteristics of extract, namely total phenols of 21.4868±3.6169 mg GAE/g dw and total flavonoids of 26.0382±5.69773 mg QE/g dw. The verification results showed that total phenols of 12.93451±1.425 mg GAE/g dw and total flavonoids of 21.79954±0.9847 mg QE/g dw. Verification results do not match predictions as indicated by a p-value of less than 0.005, indicating that the model does not accurately predict the actual results.
