Abstract

Pendahuluan: Menjaga kesehatan gigi dan mulut merupakan hal yang penting di masa pandemi Covid-19 karena kesehatan gigi dan mulut yang baik akan meminimalisir pintu masuk (portal of entry) mikroorganisme. Tanaman jarak cina (Jatropha multifida Linn.) memiliki efek antiseptik yang berpotensi sama baik dengan povidone iodine yang terbukti dapat membunuh 99,99% virus SARS-CoV-2 dalam waktu 15 detik. Tujuan: untuk mengetahui potensi kandungan senyawa metabolit sekunder terpenoid getah jarak cina sebagai kandidat senyawa obat kumur alami terhadap ACE-2 – SARS-Cov-2 dalam pencegahan Covid-19 melalui uji in silico. Metode: penelitian dilakukan dengan membuat formulasi obat kumur getah tanaman jarak cina dibuat dengan konsentrasi 1%, 3%, dan 5% yang kemudian dilakukan evaluasi sediaan yang meliputi uji organoleptis, homogenitas, pH, dan viskositas. Penelitian ini menggunakan metode in silico dengan menggunakan software Molegro Virtual Docker 5 licenced sebagai molecular docking, Biovia Discovery Studio ver 21.1.1, dan Pymol sebagai visualisasinya. Hasil: senyawa metabolit sekunder terpenoid dalam getah tanaman jarak cina (Jatropha multifida Linn.), yaitu multifidone, multidione, multifolone, multifidanol, multifidenol, dan iodine memiliki energi ikatan terhadap Protein Spike SARS-CoV-2 reseptor ACE2 lebih rendah daripada povidone iodine. Kesimpulan: Kandungan terpenoid pada getah tanaman jarak cina (Jatropha multifida Linn.) berpotensi sebagai terapi preventif dan kuratif Covid-19.

English Abstract

Introduction: Maintaining oral and dental health is important during the Covid-19 pandemic because good oral and dental health will minimize the portal of entry for microorganisms. Chinese jatropha (Jatropha multifida Linn.) has an antiseptic effect that is potentially as good as povidone iodine which has been shown to kill 99.99% of the SARS-CoV-2 virus within 15 seconds. Objective: To determine the potential content of terpenoid secondary metabolites of Chinese jatropha gum as a candidate for natural mouthwash compounds against ACE-2 – SARS-Cov-2 in the prevention of Covid-19 through an in silico test. Methods: The research was conducted by making a mouthwash formulation of Chinese jatropha gum with a concentration of 1%, 3%, and 5% which was then evaluated for the preparations which included organoleptic, homogeneity, pH, and viscosity tests. This research uses in silico method using Molegro Virtual Docker 5 licensed software as molecular docking, Biovia Discovery Studio ver 21.1.1, and Pymol as visualization. Results: terpenoid secondary metabolites in the sap of the Chinese jatropha plant (Jatropha multifida Linn.), namely multifidone, multidione, multifolone, multifidanol, multifidenol, and iodine have lower binding energies to the SARS-CoV-2 ACE2 receptor Spike Protein than povidone iodine. Conclusion: The terpenoid content in the sap of the Chinese jatropha plant (Jatropha multifida Linn.) has the potential as a preventive and curative therapy for Covid-19.
