Abstract

Tujuan penelitian ini adalah mengetahui respon pertumbuhan tunas stevia dengan penambahan hormon thidiazuron (TDZ) dan benzyl amino purine (BAP) masing-masing dengan konsentrasi 0,5; 1 dan 2 mg/L dan mengetahui perbandingan efektivitas penggunaan TDZ dan BAP terhadap induksi tunas tanaman stevia. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) 2 faktorial dengan variasi jenis Zat Pengatur Tumbuh (ZPT) dan variasi konsentrasi ZPT yaitu 0,5; 1 dan 2 mg/L yang ditambahkan pada media dasar Murashige dan Skoog (MS), sehingga terdapat 7 perlakuan, masingmasing 5 ulangan. Eksplan nodus disterilisasi dan dipotong 2 cm, kemudian diinokulasikan pada medium tanpa penambhan ZPT sebagai kontrol (T0B0), T0,5 ; T1, T2, B0,5 ; B1, dan B2. Setiap botol kultur berisi 3 eksplan. Hasil yang diperoleh terdapat perbedaan nyata pada jumlah tunas, tinggi tunas, jumlah daun dan berat eksplan, sementara waktu muncul tunas dan jumlah nodus tidak berbeda nyata. Jumlah tunas terbanyak dihasilkan oleh perlakuan BAP 2 mg/L (B2) dan terendah adalah perlakuan TDZ 1 mg/L (T1). Tinggi tunas terbaik ditunjukkan oleh perlakuan T0B0 dan terendah adalah perlakuan BAP 0,5 mg/L (B0,5). Jumlah daun tertinggi dihasilkan oleh perlakuan TDZ 1 mg/L (T1) dan BAP 1 mg/L (B1) sementara, jumlah daun terendah dihasilkan oleh perlakuan BAP 2 mg/L (B2). Berat eksplan bertunas tertinggi dihasilkan TDZ 1 mg/L (T1) dan terendah pada BAP 2 mg/L (B2). Hormon BAP lebih efektif dalam pertumbuhan jumlah tunas, sementara hormon TDZ lebih efektif dalam pertumbuhan tinggi tunas, jumlah daun dan berat eksplan.

English Abstract

The aim of this research was to determine the growth response of stevia shoots with the addition of the hormone thidiazuron (TDZ) and benzyl amino purine (BAP) each at a concentration of 0.5; 1 and 2 mg/L and to determine the comparative effectiveness of using TDZ and BAP on the induction of stevia plant shoots. This research used a 2 factorial Completely Randomized Design (CRD) with variations in the type of Growth Regulator (GR) and variations in GR concentration, namely 0.5; 1 and 2 mg/L were added to Murashige and Skoog (MS) basic media, so that there were 7 treatments, 5 replications each. The 2 cm sterilized- node explants inoculated in medium without the addition of ZPT as a control (T0B0), T0.5; T1, T2, B0.5 ; B1, and B2. Each bottle contains 3 explants. The results showed significant differences in the number of shoots, shoot height, number of leaves and sproued-explant weight, while the time of shoot emergence and number of nodes were not significantly different. The highest number of shoots was produced by the 2 mg/L BAP treatment (B2) and the lowest was the 1 mg/L TDZ treatment (T1). The control has the highest shoot, while in the BAP 0,5 mg/L (B0,5) treatment has the lowest. The highest number of leaves was produced by the TDZ 1 mg/L (T1) and BAP 1 mg/L (B1) treatment, while the lowest number of leaves was produced by the BAP 2 mg/L (B2) treatment. The highest weight of germinated explants was produced by TDZ 1 mg/L (T1) and the lowest was BAP 2 mg/L (B2). The BAP hormone was more effective in growing the number of shoots, while the TDZ hormone was more effective in growing shoot height, number of leaves and explant weight.
