Abstract

Pemeriksaan hematologi pada Buaya Muara (Crocodylus porosus) berupa perhitungan nilai total eritrosit, kadar hemoglobin dan nilai PCV yang dapat digunakan untuk deteksi kondisi kesehatan. Beberapa faktor kesalahan yang dapat mempengaruhi hasil pemeriksaan hematologi seperti, jauhnya jarak pengambilan sampel menuju fasilitas pemeriksaan hematologi, waktu yang ditempuh dalam pengambilan darah, serta suhu dan lama penyimpanan sampel darah. Penelitian ini bertujuan untuk mengetahui perbedaan nilai total eritrosit, kadar hemoglobin dan nilai Packed Cell Volume (PCV) darah Buaya Muara yang dipengaruhi oleh lama penyimpanan. Spesimen darah dikoleksi dari 30 ekor buaya yang masing-masing sebanyak 1 mL dan disimpan dalam tabung lithium heparin dengan suhu penyimpanan 4-6ºC. Pemeriksaan hematologi dilakukan segera setelah koleksi sampel untuk mendapatkan nilai dasar dan dilakukan pemeriksaan 24 jam sekali selama 96 jam. Hasil perhitungan nilai total eritrosit pada jam ke-0 (kontrol) sebesar 0,33±0,28x106 sel/µL; jam ke-24, 0,25±0,17x106 sel/µL; jam ke-48, 0,22± 0,10x106 sel/µL; jam ke-72, 0,20±0,09x106 sel/µL dan jam ke-96, 0,20±0,10x106 sel/µL. Penurunan signifikan (P<0.05) terjadi pada perhitungan total eritrosit jam ke-24 hingga jam ke-96. Hasil perhitungan kadar hemoglobin pada jam ke-0 (kontrol) 13,0±3,6 g/dL; jam ke-24, 12,9±3,5 g/dL; jam ke-48, 13,1± 3,7 g/dL; jam ke-72 13,5±4,7 g/dL dan jam ke-96 14,7 ±4,8 g/dL. Kenaikan signifikan (P>0,05) dari jam ke-48 hingga jam ke-96. Hasil perhitungan nilai packed cell volume pada jam ke-0 (kontrol), 37,8±13,2%; jam ke-24, 38,6±14,9%; jam ke-48, 36±14,9%; jam ke-72, 37,7±14,5% dan jam ke-96, 35,9±15,3%. Tidak terjadi penurunan signifikan (P>0,05) pada nilai packed cell volume. Kesimpulan penelitian ini yakni terjadi penurunan nilai total eritrosit, kenaikan kadar hemoglobin dan tidak ada penurunan nilai PCV yang dipengaruhi oleh lama penyimpanan.

English Abstract

Estuarine crocodile (Crocodylus porosus) blood can be used as a parameter in calculating total erythrocytes, hemoglobin levels and PCV values for testing to detect the patient's condition. Sample handling includes challenges in hematology examination such as storage time which can influence several factors in the results of hematology testing. This study aims to determine the comparison of total erythrocyte values, hemoglobin levels and packed cell volume (PCV) values in estuarine crocodile blood specimens which are influenced by storage time. 30 samples of 150 mL of crocodile blood were collected and taken via the coccygeal vein. which is stored in a lithium heparin tube and immediately carried out manual inspection at the 0th hour every 24 hours for 96 hours. The calculation results of the total erythrocyte value at hour 0 (control) were 0.33 ± 0.28x106 cells/µL; 24th hour, 0.25±0.17x106 cells/µL; 48th hour, 0.22 ± 0.10x106 cells/µL; 72nd hour, 0.20±0.09x106 cells/µL and 96th hour, 0.20±0.10x106 cells/µL. A significant decrease (P<0.05) occurred in total erythrocyte counts from 24 to 96 hours. The results of the calculation of hemoglobin levels at hour 0 (control) were 13.0 ± 3.6 g/dL; 24th hour, 12.9±3.5 g/dL; 48th hour, 13.1 ± 3.7 g/dL; 72nd hour 13.5 ± 4.7 g/dL and 96th hour 14.7 ± 4.8 g/dL. The increase was significant (P>0.05) from the 48th hour to the 96th hour. Calculation results of packed cell volume values at hour 0 (control), 37.8 ± 13.2%; 24th hour, 38.6±14.9%; 48th hour, 36±14.9%; 72nd hour, 37.7±14.5% and 96th hour, 35.9±15.3%. There was no significant decrease (P>0.05) in the packed cell volume value. The conclusion of this study was that there was a decrease in the total value of erythrocytes, an increase in hemoglobin levels and there was no decrease in the value of PCV which was influenced by storage time.
