Abstract

Penelitian ini bertujuan untuk menganalisis pengaruh kombinasi auksin (2,4-D atau NAA) dan sitokinin (BA) terhadap pembentukan kalus serta tekstur kalus kurma (Phoenix dactylifera L. var. Ajwa) secara in vitro. Biji kurma dikecambahkan secara in vitro dan radikula umur 23-28 HSS digunakan sebagai eksplan untuk induksi kalus. Induksi kalus dilakukan pada media MS dengan penambahan ZPT berupa auksin yaitu 2,4-D atau NAA (5, 10, dan 15 mg/L) yang dikombinasikan dengan BA 1 mg/L. Tiga buah eksplan diinokulasikan pada media dan eksplan diinkubasi pada suhu 25 ± 0,2 ºC. Tiap perlakuan terdapat lima ulangan.Kecambah dihitung terkait persentase berkecambah. Respons eksplan diamati setiap hari selama 12 minggu terkait persentase eksplan tidak kontaminasi dan tidak browning (%), saat munculnya kalus (hari setelah inokulasi (HSI)), persentase eksplan membentuk kalus (%), berat basah kalus (g), serta morfologi (warna dan tekstur) kalus. Data kualitatif dianalisis secara deskriptif, sedangkan data kuantitatif dianalisis secara statistik dengan Analysis of Variance (ANOVA) satu arah dengan selang kepercayaan 95% menggunakan software IBM SPSS Statistics 27.0. Hasil penelitian menunjukkan bahwa kalus terbentuk pada perlakuan D5B1 (5 mg/L 2,4-D dan 1 mg/L BA) pada umur 0,94 HSI; N10B1 (10 mg/L NAA dan 1 mg/L BA) pada umur 2,14 HSI; serta N15B1 (15 mg/L NAA dan 1 mg/L BA) pada umur 10,2 HSI. Persentase eksplan tidak kontaminasi dan tidak browning tertinggi (100%) pada perlakuan N15B1. Persentase eksplan membentuk kalus tertinggi (13,34%) pada perlakuan N15B1. Kalus yang terbentuk memiliki tekstur remah dan kompak. Warna kalus pada perlakuan D5B1 adalah putih kekuningan, N10B1 berwarna cokelat, dan N15B1 berwarna putih kekuningan.

English Abstract

This research aims to analyze the effect of auxin (2,4-D or NAA) with cytokinin (BA) combination on callus formation and callus texture of date palm (Phoenix dactylifera L. var. Ajwa) in vitro. Date palm seeds are germinated in vitro and radicles that grown 23-28 days after sowing were used as explants for callus induction. Callus induction was carried out on MS media with the addition of PGR in the form of auxin (2,4-D or NAA at 5, 10, and 15 mg/L) combined with BA at 1 mg/L. Three explants were inoculated on the media and the explants were incubated at 25 ± 0.2 ºC. Each treatment had five replications. Seeds was observed related to percentage of seeds germination (%). Explant responses was observed every day for 12 weeks related to percentage of explants with no contamination and no browning (%), callus emergence (days after inoculation (DAI)), percentage of explants forming callus (%), callus fresh weight (g), and callus morphology (color and texture). Qualitative data was analyzed descriptively, while quantitative data was analyzed statistically using one-way Analysis of Variance (ANOVA) with 95% confidence interval using IBM SPSS Statistics 27.0 software. Results showed that callus was formed in D5B1 treatment (5 mg/L 2,4-D and 1 mg/L BA) at the age of 0,94 DAI; N10B1 (10 mg/L NAA and 1 mg/L BA) at the age 2,14 DAI; and N15B1 (15 mg/L NAA and 1 mg/L BA) at the age 10,2 DAI. The highest percentage of explants with no contamination and no browning (100%) was in N15B1 treatment. The highest percentage of explants forming callus (13.34%) in N15B1. The callus formed has a friable and compact texture. The color of the callus in D5B1 treatment is yellowish white, N10B1 is brown, and N15B1 is yellowish white.
