Abstract

Air merupakan bagian penting dari semua jenis kehidupan karena berperan sangat penting. Air digunakan untuk berbagai macam kegiatan seperti minum, pertanian, industri, perikanan dan rekreasi, salah satu contohnya adalah dalm pembuatan es batu. Maka dari itu, kebersihan air perlu diperhatikan agar tidak menyebabkan penyakit seperti water-borne disease. Water-borne disease adalah penyakit yang ditularkan ke manusia karena adanya kontaminasi mikroorganisme patogen atau zat-zat berbahaya yang terdapat di dalam air. Oleh karena itu dilakukan upaya pencegahan terhadap water-borne disease yang dilakukan dengan mendeteksi mikroba patogen penyebab water-borne disease pada air yang digunakan untuk produksi es batu pada jajanan tepatnya di 45 Sekolah Dasar Negeri Kecamatan Lowokwaru Kota Malang mengingat belum adanya regulasi standar kebersihan es batu. Salmonella spp. dan Vibrio cholerae merupakan target mikroba patogen dari penelitian karena kedua patogen tersebut bersifat stabil dalam kondisi perairan dan berpotensi untuk menyebabkan penyakit berbahaya. Sampel air es batu dari 20 SD diambil berdasarkan multistage sampling yaitu stratified random sampling. Deteksi mikroba patogen pada sampel akan menggunakan metode Polymerase Chain Reaction (PCR) karena dalam pelaksanaannya sederhana dan dapat dengan cepat dilihat hasilnya. Penelitian akan dimulai dengan pengambilan dan preparasi sampel, isolasi DNA, uji konsentrasi dan kemurnian DNA, ampifikasi DNA, elektroforesis, dan diakhiri dengan mengamati pita yang terbentuk saat visualisasi DNA. Setelah dilaksanakan deteksi PCR, ditemukan hasil bahwa sebanyak 100% dari sampel es batu terdapat keberadaan bakteri Salmonella spp., dan sekitar 15% sampel es terdapat keberadaan bakteri Vibrio cholerae berdasarkan deteksi yang dilaksanakan.

English Abstract

Water is an important part of all types of life because it plays a very important role. Water is used for various activities such as drinking, agriculture, industry, fishing, and recreation, one example is in making ice cubes. Therefore, it is necessary to pay attention to the cleanliness of the water so that it does not cause diseases such as water-borne diseases. Water-borne disease is a disease that is transmitted to humans due to contamination by pathogenic microorganisms or dangerous substances contained in water. Therefore, efforts are being made to prevent water-borne disease by detecting pathogenic microbes that cause water-borne disease in the water used for the production of ice cubes in snacks, to be precise at 45 State Elementary Schools in Lowokwaru District, Malang City, considering that there are no standard regulations for the cleanliness of ice cubes. Salmonella spp. and Vibrio cholerae are the target microbial pathogens for research because both pathogens are stable in aquatic conditions and have the potential to cause dangerous diseases. Ice cube water samples from 20 elementary schools were taken based on multistage sampling, namely stratified random sampling. Detection of pathogenic microbes in samples will use the Polymerase Chain Reaction (PCR) method because the implementation is simple and the results can be seen quickly. The research will begin with sample collection and preparation, DNA isolation, DNA concentration and purity testing, DNA amplification, electrophoresis, and ends by observing the bands formed during DNA visualization. After PCR detection was carried out, the results were found that as many as 100% of the ice cube samples contained the presence of Salmonella spp. bacteria and around 15% of the ice samples contained the presence of Vibrio cholerae bacteria based on the detection carried out.
