Abstract

Hipofungsi ovarium merupakan suatu gangguan sistem reproduksi pada ternak yang menyebabkan ternak mengalami penurunan fungsi ovarium. Tujuan dari penelitian ini yaitu untuk mengembangkan hewan mencit model hipofungsi ovarium dengan induksi LPS (lipopolisakarida) dan antagonis GnRH Ganirelix acetate serta mengamati perubahan gambaran histopatologi organ uterus mencit dan ekspresi reseptor estrogen α pada uterus. Prosedur penelitian ini meliputi persiapan hewan coba dan aklimatisasi selama 7 hari, perlakuan induksi hipofungsi ovarium, koleksi organ, pembuatan dan pengamatan preprarat histopatologi organ uterus, pengamatan ekspresi reseptor α metode imunohistokimia serta analisis data. Pada penelitian ini menggunakan tiga kelompok perlakuan yang terdiri atas enam ekor mencit Swiss Webster betina, berusia ≥ 4 bulan, berat badan 20-30gram. Kelompok perlakuan tersebut yakni kelompok K- (kontrol negatif), kelompok P1 (injeksi lipopolisakarida 0.0115mg/mencit) dan kelompok perlakuan P2 (injeksi Ganirelix acetate 0.00195mg/mencit. Penentuan dosis lipopolisakarida pada penelitian menggunakan dosis low dose mencit pada jurnal (Lv et al., 2021), sedangkan dosis Ganirelix acetate menggunakan konversi dosis yang mengacu pada jurnal (Wilcox et al., 2005) dan (Firmawati et al., 2019). Histopatologi uterus diamati dengan mengukur ketebalan endometrium serta diamati ekspresi reseptor estrogen α dengan metode imunohistokimia. Dilakukan analisis data dengan uji one way ANOVA (α = 0.05) dan hasil menunjukkan tidak ada perbedaan yang signifikan antar kelompok perlakuan, baik pada ketebalan endometrium maupun pada ekspresi reseptor estrogen α uterus mencit.

English Abstract

Ovarian dysfunction is a disorder of the reproductive system in livestock that leads to decreased ovarian function in livestock.The purpose of this study was to develop a mouse model of ovarian dysfunction caused by lipopolysaccharide and GnRH antagonist, and observe changes in the histopathological picture of mouse uterus and estrogen receptor expression α in the uterus.This research protocol includes preparing experimental animals and acclimating them for 7 days, induction treatment for ovarian dysfunction, organ harvesting, observing histopathological preparate of the organs uterus, while estrogen receptor α expression is observed by immunohistochemistry. This study used three treatment groups, each consisting of six female Swiss Webster rats, ≥4 months old and weighing 20 to 30 grams. The treatment groups were negative control (K-) group, group P1 (lipopolysaccharide 0.0115 mg/mouse) and group P2 (Ganirelix acetate 0.00195 mg/mouse). Lipopolysaccharide dose determination in the study used low-dose lipopolysaccharide to mice in the review (Lv et al., 2021), while Ganirelix acetate dosing used a dose converter referencing the reviews (Wilcox et al., 2005) and ( Firmawati et al., 2019). Histopathological features of the uterus were observed by examining and measuring changes in endometrial thickness and estrogen receptor α expression by immunohistochemistry. Then, data analysis was performed using one-way ANOVA (α = 0.05) test to determine the treatment effect in each group. The results showed that there were no significant differences between treatment groups, either in endometrial thickness or in the expression of estrogen receptor α in the mouse uterus.
