Abstract

Beberapa uji hematologi seperti, perhitungan total eritrosit, kadar hemoglobin dan indeks eritrosit dapat digunakan untuk menilai kondisi pasien. Faktor kesalahan seperti, pengananan sampel, pengangkutan, suhu penyimpanan, pemrosesan yang tertunda dan durasi penyimpanan dapat merubah kualitas hasil uji hematologi Penelitian ini bertujuan mengetahui perbedaan pada total eritrosit, kadar hemoglobin dan indeks eritrosit pada spesimen darah Iguana hijau (Iguana iguana) yang dipengaruhi durasi penyimpanan pada suhu lemari pendingin (4oC) selama 96 jam. Spesimen darah dikoleksi dari 19 ekor Iguana hijau dewasa yang disimpan di dalam tabung lithium heparin. Pemeriksaan hematologi dilakukan segera setelah koleksi sampel untuk mendapatkan nilai dasar dan dilakukan pemeriksaan 24 jam sekali selama 96 jam. Hasil perhitungan total eritrosit pada jam ke-0 (kontrol) sebesar 1,0±0,4 x 106 sel/μL; jam ke-24, 0,8±0,5 x 106 sel/μL; jam ke-48, 0,7±0,3 x106 sel/μL; jam ke-72, 0,6±0,4 x 106 sel/μL; jam ke-96, 0,4±0,3 x 106 sel/μL. Penurunan signifikan (p<0,05) terjadi pada perhitungan total eritrosit jam ke-72 dan jam ke-96. Hasil perhitungan kadar hemoglobin pada jam ke-0 (kontrol) ; 10,5±2,5 g/dL; jam ke-24, 10,4 ± 2,7 g/dL; jam ke-48, 9,9 ± 2,2; jam ke-72, 9,6±3,8 g/dL; jam ke-96 8,8±3,0 g/dL. Penurunan terjadi dari jam-0 hingga jam ke-96 namun tidak secara signifikan (p>0,05). Hasil perhitungan MCV pada jam ke-0 (kontrol) 565±334 fL; jam ke-24, 677±448 fL; jam ke-48, 785±867 fL, jam ke-72, 493±428 fL; jam ke-96, 1194±3420. Pada nilai MCV tidak terjadi penurunan. Hasil perhitungan nilai MCH pada jam ke-0 (kontrol), 130±61 pg; jam ke-24, 163±86 pg; jam ke-48, 197±166 pg, jam ke-72, 279±305 pg, jam ke-96, 1020±1442 pg. Terjadi kenaikan pada nilai MCH. Hasil perhitungan nilai MCHC pada jam ke-0 (kontrol) 25,4±7,0 g/dL; jam ke-24, 26,5±10,0 g/dL; jam ke-48, 25,6±9,3 g/dL; jam ke-72, 23,4±19,0 g/dL; jam ke-96, 19,8±17,5 g/dL. Pada nilai MCHC tidak terjadi penurunan secara signifikan pada jam ke-0 hingga jam ke-96. Kesimpulan penelitian ini adalah adanya perbedaan berupa penurunan nilai total eritrosit dan nilai Hb namun tidak terjadi penurunan pada nilai indeks eritrosit, yang dipengaruhi oleh durasi penyimpanan.

English Abstract

Several hematology tests such as total erythrocyte count, hemoglobin level and erythrocyte index can be used to assess the patient's condition. Error factors such as sample handling, transportation, storage temperature, delayed processing and storage duration can change the quality of hematology test results. This study aims to determine differences in total erythrocytes, hemoglobin levels and erythrocyte index in green Iguana (Iguana iguana) blood specimens as affected by storage duration at refrigerator temperature (4o C) for 96 hours. Blood specimens were collected from 19 adult green Iguanas stored in lithium heparin tubes. Hematological examinations were performed immediately after sample collection to obtain baseline values and examined every 24 hours for 96 hours. The total erythrocyte count at hour 0 (control) was 1.0±0.4 x 106 cells/μL; hour 24, 0.8±0.5 x 106 cells/μL; hour 48, 0.7±0.3 x106 cells/μL; hour 72, 0.6±0.4 x 106 cells/μL; hour 96, 0.4±0.3 x 106 cells/μL. A significant decrease (p<0.05) occurred in the calculation of total erythrocytes at 72nd hour and 96th hour. Hemoglobin levels were calculated at hour 0 (control); 10.5 ± 2.5 g/dL; hour 24, 10.4 ± 2.7 g/dL; hour 48, 9.9 ± 2.2; hour 72, 9.6 ± 3.8 g/dL; hour 96, 8.8 ± 3.0 g/dL. The decrease occurred from hour-0 to hour-96 but not significantly (p>0.05). The results of MCV calculation at hour 0 (control) 565 ± 334 fL; hour 24, 677 ± 448 fL; hour 48, 785 ± 867 fL, hour 72, 493 ± 428 fL; hour 96, 1194 ± 3420. There was no decrease in the MCV value. The results of the calculation of MCH values at hour 0 (control), 130±61 pg; hour 24, 163±86 pg; hour 48, 197±166 pg, hour 72, 279±305 pg, hour 96, 1020±1442 pg. There was an increase in the MCH value. The results of the calculation of MCHC values at hour 0 (control) 25.4 ± 7.0 g/dL; hour 24, 26.5 ± 10.0 g/dL; hour 48, 25.6 ± 9.3 g/dL; hour 72, 23.4 ± 19.0 g/dL; hour 96, 19.8 ± 17.5 g/dL. The MCHC value did not decrease significantly from the 0th hour to the 96th hour. The conclusion of this study is that there is a difference in the form of a decrease in the value of total erythrocytes and Hb values but there is no decrease in the value of the erythrocyte index, which is influenced by the duration of storage.
