Abstract

Tubuh memiliki mekanisme pertahanan alami berupa antioksidan endogen. Adapun enzim antioksidan endogen tersebut adalah Superoxide Dismutase (SOD) yang berfungsi untuk mengatur kadar Reactive Oxygen Species (ROS) yang meningkat kadarnya dalam tubuh saat keadaan stres dan inflamasi. Induksi stres menimbulkan ketidakseimbangan antara produksi ROS dengan kadar SOD jaringan hepar karena respon stres oksidatif. Penelitian sebelumnya melaporkan antosianin ubi jalar ungu dapat menurunkan kadar SOD tikus aterogenik. Penelitian terkait manfaat ANT ubi jalar ungu masih terbatas, (Ipomoea batatas L.) memiliki manfaat sebagai antioksidan eksogen alami untuk mengatasi stres fisik dan psikologis. Oleh karena itu, penelitian ini bertujuan untuk menganalisis pemberian ANT ubi jalar ungu terhadap kadar SOD hepar mencit yang dipapar stres fisik dan psikologis. Dalam eksperimen ini, mencit jantan dewasa usia 6-8 minggu dipaparkan pada dua jenis stres: stres dan stres psikologis selama 14 hari dengan durasi 2 jam per hari dengan jadwal acak. Mencit dibagi menjadi 5 kelompok perlakuan yaitu kontrol, stres, stres + ANT 10mg/kgBB, stres + ANT 20mg/kgBB, dan stres + ANT 40mg/kgBB. Pengukuran kadar SOD jaringan hepar digunakan sebagai indikator respons fisiologis. Hasil menunjukkan bahwa paparan terhadap stres fisik dan psikologis menyebabkan peningkatan dalam kadar SOD jaringan hepar secara signifikan (p=0,013). Peningkatan kadar SOD secara signifikan pada kelompok yang diinduksi stres, dan ketiga kelompok yang diinduksi stres + antosianin memiliki kadar SOD lebih rendah dari kelompok stres meski tidak signifikan sehingga ekstrak antosianin potensial sebagai antioksidan. Untuk penelitian yang selanjutnya diharapkan dapat meneliti dosis antosianin yang lebih tinggi dan optimasi durasi terapi yang lebih lama pada mencit.

English Abstract

A human body naturally possesses endogenous antioxidants as a defense mechanism. One such endogenous antioxidant enzyme is Superoxide Dismutase (SOD), which functions to regulate the levels of Reactive Oxygen Species (ROS) that increase during periods of stress and inflammation in the body. The induction of stress creates an imbalance between ROS production and the SOD levels in the hepatic tissues due to the oxidative stress response. Previous studies have reported that anthocyanins from purple sweet potatoes can reduce the levels of SOD in atherogenic rats. The research on the benefits of purple sweet potato anthocyanins (ANT) is still limited; however, (Ipomoea batatas L.) is known to have natural exogenous antioxidant properties to alleviate physical and psychological stress. Therefore, the aim of this study is to analyze the administration of purple sweet potato ANT on the levels of hepatic SOD in mice exposed to physical and psychological stress. In this experiment, adult male mice aged 6-8 weeks were exposed to two types of stress: physical stress and psychological stress for 14 days, with a duration of 2 hours per day on a random schedule. The mice were divided into 5 treatment groups: control, stress, stress + ANT 10mg/kgBW, stress + ANT 20mg/kgBW, and stress + ANT 40mg/kgBW. Measurement of hepatic SOD levels was used as an indicator of physiological response. The results indicate that exposure to physical and psychological stress significantly increases the levels of hepatic SOD (p=0.013). There was a significant increase in SOD levels in the stress-induced group, and the three groups induced with stress + anthocyanin had lower SOD levels than the stress group, although not statistically significant. This suggests that anthocyanin extract has the potential as an antioxidant. Subsequent research is expected to explore higher anthocyanin doses and optimize the duration of therapy in mice.
