Abstract
Seleksi berbasis marka genetik salah satu upaya untuk meningkatkan kualitas genetik secara cepat dan akurat. Asosiasi polimorfisme gen growth hormone (GH) dengan sifat-sifat kuantitatif seperti: bobot badan, lingkar dada, panjang badan, dan tinggi badan pada domba Peranakan Texel berperan penting dalam program seleksi berbasis marka genetik (MAS). Tujuan penelitian untuk mengkaji keragaman genetik dan asosiasi gen GH dengan sifat-sifat kuantitatif domba Peranakan Texel. Materi yang digunakan yaitu domba Peranakan Texel sebanyak 42 ekor betina. Metode penelitian adalah metode survei, pengambilan sampel dipilih secara purposive sampling, sampel darah dikoleksi dari peternakan Agri Lestari Dairy Farm. Gen GH memiliki panjang 661 bp. Fragment DNA diampflifikasi dan diidentifikasi genotipe menggunakan metode PCR-RFLP dengan enzim restriksi MspI dan AluI. Assosiasi polimorfisme gen GH dengan sifat-sifat kuantitatif dianalisis menggunakan analysis of variance. Hasil penelitian menunjukkan gen GH-AluI bersifat monomorfik, ditemukan satu genotipe gen GH-AluI (GG) dan alel (G). Sedangkan gen GH-MspI bersifat polimorfik dengan ditemukan SNP pada posisi c.573 G>A. Nilai PIC sebesar 49,98% menunjukkan polimorfisme sedang pada lokus gen GH. Keseimbangan Hardy-Weinberg menunjukkan populasi domba Peranakan Texel dalam keadaan seimbang. Ditemukan tiga genotipe gen GH-MspI (AA, AG, GG) dan dua alel (A dan G). Frekuensi genotipe AA, AG, GG masing-masing 0.24, 0.55, 0.21, sedangkan frekuensi alel A dan G masing-masing 0.51 dan 0.49. Hasil analysis of variance menunjukkan bahwa terdapat asosiasi antara keragaman gen GH-MspI yang ditemukan dengan sifat-sifat kuantitatif (p<0,01).
English Abstract
Selection based on genetic markers is one of the efforts to improve genetic quality quickly and accurately. The association of growth hormone (GH) gene polymorphisms with quantitative traits such as body weight, chest girth body length, and body height in Texel-breed sheep is an important role in the marker-assisted selection program (MAS). The research aimed to study genetic diversity and the association of GH genes with quantitative traits of Texel-breed sheep. The material used is 42 ewes. The research method is a survey method, the sample was selected by purposive sampling, and blood samples were collected from the Agri Lestari Dairy Farm. The GH gene is 661 bp long. DNA fragments were amplified and the genotype was identified using the PCR-RFLP method with restriction enzymes MspI and AluI. The association of GH gene polymorphisms with quantitative traits was analyzed using analysis of variance. The results showed that the GH-AluI gene was monomorphic, one genotype was found for the GH-AluI gene (GG) and allele (G). While the GH-MspI gene is polymorphic, SNP was found at position c.573 G>A. The PIC value is 49.98% indicates moderate polymorphism at the GH gene locus. The Hardy-Weinberg equilibrium shows that the Texel-breed sheep population is in balance. The genotype frequencies of AA, AG and GG were 0.24, 0.55, and 0.21, respectively, while the frequencies of the A and G alleles were 0.51 and 0.49, respectively. The results of the analysis of variance showed that there was an association between the diversity of the GH-MspI gene found with quantitative traits (p<0.01).
