Abstract

Diabetes Mellitus (DM) adalah penyakit metabolik yang berhubungan dengan sekresi insulin yang ditandai dengan hiperglikemia dan gangguan metabolisme karbohidrat, lemak, dan protein, karena kekurangan insulin, resistensi insulin, atau keduanya. Salah satu obat diabetes melitus 2 adalah acarbose. Penggunaan obat jenis ini memiliki efek samping yang dapat menyebabkan diare dan kembung dan harus dihindari bagi pasien dengan gangguan ginjal. Dari efek samping itu dicari bahan alam yang berfungsi menghambat α-glukosidase salah satunya yaitu mangrove. Mangrove lindur (B. gymnorrhiza) adalah salah satu jenis mangrove yang memiliki aktivitas biologi sebagai antimikroba karena mengandung senyawa fitokimia, seperti: steroid, triterpenoid, saponin, flavonoid, alkaloid dan tanin. Tujuan umum dari penelitian ini yaitu untuk mengetahui potensi fraksi metanol daun lindur (B. gymnorrhiza) sebagai inhibitor enzim α-glukosidase. Tujuan khusus dari dilakukannya penelitian ini yaitu untuk mengidentifikasi senyawa fitokimia yang terkandung dalam fraksi metanol daun lindur (B. gymnorrhiza) dan untuk mengetahui potensi dari senyawa fitokimia dalam fraksi metanol daun lindur (B. gymnorrhiza) sebagai inhibitor enzim α-glukosidase. Mekanisme kerja inhibitor α-glukosidase adalah dengan cara menghambat pemecahan karbohidrat dari makanan menjadi glukosa untuk mengendalikan kadar gula darah. Inhibitor α-glukosidase menghambat aktivitas α-glukosidase dengan menempati tempat pengikatan α-glukosidase dan gula secara reversibel, sehingga mengurangi degradasi polisakarida, menunda penyerapan karbohidrat di usus, dan mencapai efek hipoglikemik Analisis LC-MS/MS dilakukan dilakukan di Puslabafor Bogor pada tanggal 12 Juni 2023. Kemudian dilakukan Identifikasi senyawa fitokimia daun mangrove B. gymnorrhiza menggunakan software Masslynx Waters versi 4.1. dan didapatkan beberapa jenis senyawa fitokimia yang dihasilkan seperti pada golongan flavonoid yaitu chrysoeriol 7-o-glucoside, isorhamnetin, quercetin, tricin, hispidulin dan glycitein. Diperoleh senyawa golongan lipid yaitu LPC 18:3, LPC 16:0 dan 1-Stearoyl-sn-glycero-3-phosphocholine dengan nilai total flavonoid yaitu 12416,91 mg QE/g. Nilai IC50 dari fraksi metanol daun lindur (B. gymnorrhiza) yaitu 6242,44 ppm, sedangkan nilai IC50 dari acarbose yaitu 147,58±4,33 ppm. Fraksi metanol daun lindur (B. gymnorrhiza) memiliki aktivitas penghambatan enzim α-glukosidase sangat lemah diabnding acarbose. Dari hasil penelitian ini, perlu dilakukan penelitian lebih lanjut menggunakan fraksi yang berbeda. Selain itu dapat dilakukan penelitian lebih lanjut secara in silico dan in vivo.

English Abstract

Diabetes Mellitus (DM) is a metabolic disease related to insulin secretion characterized by hyperglycemia and disorders of carbohydrate, fat and protein metabolism characterized by hyperglycemia, or high blood sugar, due to insulin deficiency, insulin resistance, or both. One of the drugs for Diabetes mellitus 2 is acarbose. The use of this type of drug has side effects that can cause diarrhea and bloating and should be avoided for patients with kidney disorders. From these side effects, look for natural ingredients that function to inhibit α-glucosidase, one of which is mangrove. Lindur mangrove (B. gymnorrhiza) is a type of mangrove that has biological activity as an antimicrobial because it contains phytochemical compounds, such as: steroids, triterpenoids, saponins, flavonoids, alkaloids and tannins. The general aim of this research is to determine the potential of methanol extract of lindur leaves (B. gymnorrhiza) as an inhibitor of the α-glucosidase enzyme. The specific aim of this research is to identify the phytochemical compounds contained in the methanol extract of lindur leaves (B. gymnorrhiza) and to determine the potential of the phytochemical compounds in the methanol extract of lindur leaves (B. gymnorrhiza) as inhibitors of the α-glucosidase enzyme. The mechanism of action of α-glucosidase inhibitors is by inhibiting the breakdown of carbohydrates from food into glucose to control blood sugar levels. α-glucosidase inhibitors inhibit α-glucosidase activity by reversibly occupying the binding site of α-glucosidase and sugar, thereby reducing polysaccharide degradation, delaying intestinal absorption of carbohydrates, and achieving hypoglycemic effects LC-MS/MS analysis was carried out at Puslabafor Bogor on June 12 2023. Then identification of phytochemical compounds in B. gymnorrhiza mangrove leaves was carried out using Masslynx Waters version 4.1 software. and several types of phytochemical compounds were obtained, such as the flavonoid group, namely chrysoeriol 7-o-glucoside, isorhamnetin, quercetin, tricin, hispidulin and glycitein. Lipid group compounds were obtained, namely LPC 18:3, LPC 16:0 and 1-Stearoyl-sn-glycero-3-phosphocholine with a total flavonoid value of 12416.91 mg QE/g. The IC50 value of the methanol fraction of lindur leaves (B. gymnorrhiza) is 6242.44 ppm, while the IC50 value of acarbose is 147.58 ± 4.33 ppm. The methanol fraction of lindur leaves (B. gymnorrhiza) has very weak α-glucosidase enzyme inhibitory activity compared to acarbose. From the results of this research, further research needs to be carried out using different fractions. Apart from that, further research can be carried out in silico and in vivo.
