Abstract

Larutaan fiksatif terdiri dari zat kimia yang menjaga integritas jaringan setelah kematian. Larutan fiksatif juga telah digunakan untuk mnegawetkan organ untuk keperluan higienis selama transportasi jarak jauh untuk menghindari kontaminasi. Larutan fiksatif dengan bahan dasar etanol, asam asetat, dan kitosan dapat dipertimbangkan sebagai larutan fiksatif alternatif pengganti formalin yang dalam penggunaannya lebih aman bagi kesehatan. Etanol dalam larutan bertindak sebagai agen fiksatif. Kloroform ditambahkan ke dalam larutan untuk menetralkan efek penyusutan oleh etanol. Kitosan dan asam asetat berfungsi sebagai antibakteri. Kitosan juga memiliki kemampuan sebagai penghambat pertumbuhan kapang dan jamur. Tujuan penelitian ini yaitu untuk mengetahui pengaruh larutan fiksatif berbahan etanol, asam asetat, dan kitosan terhadap perubahan diameter sel aciner dan pulau langerhans pada histopatologi pankreas ayam. Dalam penelitian ini digunakan organ pankreas ayam yang dibagi menjadi 3 kelompok yang setiap kelompoknya berisi 6 organ. Pada kelompok P1 organ pankreas ayam disimpan dalam larutan fiksatif pada suhu ruang selama 3 hari sedangkan kelompok P2 organ pankreas ayam disimpan dalam larutan fiksatif pada suhu ruang selama 7 hari dan K0 sebagai control yaitu organ pankreas ayam direndam dalam larutan formalin 10% selama sehari. Pengaruh larutan fiksatif diukur secara kuantitatif dari gambaran histopatologi sel aciner dan pulau langerhans organ pankreas ayam menggunakaan mikroskop yang didukung optic lab viewer dengan software image raster dan dianalisis menggunakan aplikasi Microsoft excel dan SPSS for windows dengan metode analisis statistic menggunakan One Way ANOVA. Hasil penelitian ini menunjukkan bahwa penyimpanan organ didalam larutan etanol, asam asetat dan kitosan tidak berbeda nyata (P>0,05) antara kelompok K0, P1 dan P2.

English Abstract

Fixative solutions consist of chemicals that maintain tissue integrity after death. Fixative solutions have also been used to preserve organs for hygienic purpose during long distance transportation to avoid contamination. Fixative solutions based on ethanol, acetic acid and chitosan can be considered as an alternative fixative solution to formalin which is safer for health. Ethanol in solution acts as a fixative agent. Chloroform is added to the solution to neutralize the effects of shrinkage by ethanol. Chitosan and acetic acid function as antibacterial. Chitosan also has the ability to inhibit the growth of mold and fungus, The purpose of this study was to determine the effect of fixative solutions made from ethanol, acetic acid und chitosan on changes in the condition of aciner cells and islets of Langerhans in the histopathology of chicken pancreas. In this study chicken pancreas organs were used which were divided into 3 groups, each group containing 6 organs. In group PI the pancreatic organs were stored in fixative solution at room temperature for 3 days while in group P2 the pancreatic organs were stored in fixative solution at room temperature for 7 days and K0 store the pancreasin formalin 10% for a day. The effect of fixative solution was measured quantitatively from histopathological features of the diameter of aciner cells and Langerhans islands of chicken pancreas organs using a microscope supported by Optic Lab Viewer with raster image software and analyzed using Microsoft Excel and SPSS for windows applications with statistical analysis methods using One Way ANOVA. The results of this study indicate that the organs in a solution of ethanol, acetic acid, and chitosan was not significantly different (p>0,05) between K0, P1, and P2.
