Abstract
Pandemi COVID-19 penyebarannya masih meluas dan menimbulkan banyak kerugian. Pengembangan vaksinasi dalam upaya pencegahan COVID-19 masih diperlukan. Salah satunya yaitu penggunaan protein spike sebagai kandidat vaksin dengan sistem penghantaran live bacterial vector. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian protein spike (high conserved region (HCR) dan receptor binding domain (RBD)) terhadap persentase jumlah CD138 pada limpa mencit. Mencit dibagi ke dalam tujuh kelompok yaitu kontrol negatif(K1), vaksinasi oral L. lactis(K2), oral L. lactis pNZHCR(K3), oral L. lactis pNZRBD(K4), intranasal L. lactis(K5), intranasal L. lactis pNZHCR(K6), dan intranasal L. lactis pNZRBD(K7). Vaksinasi diberikan selama 3 hari berturut-turut dalam 2 minggu, dan dilakukan sebanyak 3 kali. Hari ke-56, mencit dikorbankan dan dilakukan isolasi limfosit dari limpa, diamati persentase jumlah CD138 dengan flowcytometry metode Fluorescence Activated Cell Sorter (FACS). Data yang didapatkan kelompok 2,3,4,5,dan 6 memiliki rata-rata persentase jumlah CD138 lebih besar daripada kontrol negatif, sedangkan kelompok 7 lebih kecil dari kontrol negatif. Hasil analisis statistik menunjukkan terdapat perbedaan yang signifikan pada kelompok vaksinasi intranasal pNZHCR dengan p < 0,05. Pemberian vaksinasi rute oral dapat meningkatkan persentase jumlah CD138 namun tidak signifikan. Dapat disimpulkan, pemberian vaksin protein spike dengan penghantaran live bacterial vector dapat menginduksi respons imun melalui peningkatan persentase jumlah CD138 di limpa mencit dengan respons terbaik pada rute intranasal. Namun, induksi respons imun belum tentu spesifik terhadap SARS-CoV-2 karena hanya mengamati respons imun secara umum, sehingga masih diperlukan penelitian lanjutan seperti menggunakan parameter imunitas seluler (seperti CD4+ dan/atau CD8+) atau percobaan paparan virus SARS-CoV-2 pada hewan coba setelah diberikan vaksin.
English Abstract
The COVID-19 pandemic is still spreading and causing a lot of damage. Vaccination development to prevent COVID-19 is still needed. One of them is the use of spike protein as a vaccine candidate with a live bacterial vector delivery system. This study aimed to determine the effect of spike protein (high conserved region (HCR) and receptor-binding domain (RBD)) on the percentage of CD138 in the spleen of mice. Mice were divided into seven groups, namely negative control (K1), oral L. lactis vaccination (K2), oral L. lactis pNZHCR (K3), oral L. lactis pNZRBD(K4), intranasal L. lactis(K5), intranasal L lactis pNZHCR(K6), and intranasal L. lactis pNZRBD(K7). Vaccination was given three consecutive days two weeks apart and carried out three times. On the 56th day, mice sacrificed, and lymphocytes were isolated from the spleen. The percentage of CD138 count was observed using flow cytometry using the Fluorescence Activated Cell Sorter (FACS) method. The data obtained by groups 2, 3, 4, 5, and 6 had an average CD138 count higher than the negative control, while group 7 was smaller than the negative control. The results of statistical analysis showed that there was a significant difference in the pNZHCR intranasal vaccination group with a p-value < 0.05. Administration of oral route of vaccination can increase the CD138 count but not significantly. The conclusion is that the administration of spike protein vaccine with live bacterial vector delivery can induce an immune response by increasing the percentage of CD138 in the spleen of mice with the best results on the intranasal route. However, the induction of immune response is not necessarily specific to SARS-CoV-2 because it only observes the general immune response, so further research is needed, such as using cellular immunity parameters (such as CD4+ or CD8+) or experimental with SARS-CoV-2 virus exposure after being vaccinated.
