Abstract

Tiap mikroorganisme yang berada di rizosfer memiliki interaksi satu sama lain. Interaksi ini dapat berupa interaksi sinergisme maupun antagonisme contohnya interaksi antara jamur arbuskula mikoriza dengan bakteri. Bakteri yang berpotensi dapat bekerja sinergis dengan mikoriza dalam meningkatkan pertumbuhan dan ketahanan tanaman adalah bakteri pelarut fosfat dan bakteri antagonis. Penelitian tentang pengaruh bakteri pelarut fosfat dan bakteri antagonis terhadap infeksi arbuskula mikoriza belum pernah dilakukan. Oleh karena itu, perlu dilakukan penelitian yang bertujuan untuk mengkaji bagaimana pengaruh bakteri antagonis dan pelarut fosfat terhadap infeksi jamur arbuskula mikoriza pada perakaran tanaman jagung. Penelitian ini dilaksanakan pada bulan November 2021 sampai Mei 2022 di Laboratorium Pengendalian Hayati I dan Rumah Kaca, Departemen Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya. Tahapan Penelitian meliputi isolasi bakteri dari media perbanyakan arbuskula mikoriza indigenous UB Forest, seleksi bakteri pelarut fosfat dan uji zona bening pelarut fosfat bakteri B5 dan B8, seleksi bakteri antagonis terhadap F. oxysporum dan uji daya hambat antagonis bakteri B5 dan B8, uji pewarnaan gram B5 dan B8 dan uji pengaruh bakteri B5 dan B8 terhadap infeksi jamur arbuskula mikoriza. Adapun variabel penelitian ini meliputi diameter zona bening bakteri pelarut fosfat, persentase daya hambat antagons terhadap F. oxysporum, dan persentase infeksi jamur arbuskula mikoriza pada perakaran tanaman jagung. Berdasarkan hasil penelitian didapatkan hasil bahwa bakteri yang didapatkan dari media perbanyakan arbuskula mikoriza adalah sebanyak 8 isolat. Dari kedelapan isolat yang merupakan bakteri pelarut fosfat dan antagonis terhadap patogen F. oxysporum adalah bakteri B5 dan B8. B5 dan B8 merupakan bakteri gram positif. Diameter zona bening pelarut fosfat B8 lebih besar dibandingkan B5 yaitu 0,33 cm sedangkan B8 adalah 0,45 cm. Persentase daya hambat yang dihasilkan B5 lebih besar dibandingkan B8 yaitu 65,45% sedangkan B8 adalah 45,07%. Persentase infeksi akar B8 pada perlakuan 10 gr arbuskula mikoriza dan 5 ml B8 berpengaruh nyata dan meningkatkan infeksi mikoriza. Sedangkan perlakuan yang lain tidak berpengaruh nyata. Berdasarkan hasil uji ini, maka bakteri pelarut fosfat dan antagonis memiliki dua pengaruh terhadap infeksi jamur arbuskula mikoriza yaitu dapat meningkatkan dan bersifat netral. Akan tetapi tidak berpengaruh negatif terhadap infeksi jamur arbuskula mikoriza.

English Abstract

Each microorganism residing in the rhizosphere has an interaction with each other. This interaction can be in the form of synergistic or antagonisms interactions, for example the interaction between mycorrhizal arbuscula fungi and bacteria. Bacteria that have the potential to work synergistically with mycorrhiza in increasing plant growth and resistance are solvent phosphate bacteria and antagonistic bacteria. Research on the influence of solvent phosphate bacteria and antagonistic bacteria on mycorrhizal arbuscular infection has never been conducted. Therefore, it is necessary to conduct research aimed at examining how the influence of antagonistic bacteria and phosphate solvents on mycorrhizal arbuscula fungal infections on the root of corn plants. This research was carried out from November 2021 to May 2022 at the Biological Control Laboratory I and Greenhouses, Department of Pests and Plant Diseases, Faculty of Agriculture, Universitas Brawijaya. The research phase included bacterial isolation from UB Forest mycorrhizal arbuscula propagation media, selection of phosphate solvent bacteria and clear zone test of bacterial phosphate solvents B5 and B8, selection of antagonistic bacteria against F. oxysporum and antagonistic inhibitory power of B5 and B8 bacteria test, gram staining test of B5 and B8, and the influence of B5 and B8 bacteria on mycorrhizal arbuscula fungal infection test. The variables of this study included the diameter of the clear zone of phosphate solvent bacteria, the percentage of inhibitory power of antagons against F. oxysporum, and the percentage of mycorrhizal arbuscular fungal infections in the root of corn plants. Based on the results of the study, the results were obtained that the bacteria obtained from the mycorrhizal arbuscula propagation media were as many as 8 isolates. Of the eight isolates that are phosphate solvent bacteria and antagonists to the pathogen F. oxysporum are bacteria B5 and B8. B5 and B8 are gram positive bacteria. The clear zone diameter of the phosphate solvent B8 is greater than that of B5 which is 0.33 cm while B8 is 0.45 cm. The percentage of inhibitory power produced by B5 is greater than B8, which is 65.45% while B8 is 45.07%. The percentage of B8 root infection on the treatment of 10 gr of mycorrhizal arbuscula and 5 ml of B8 had a noticeable effect and increased mycorrhizal infection. Meanwhile, the other treatment has no real effect. Based on the results of this test, the phosphate solvent bacteria and antagonists have two influences on mycorrhizal arbuscular fungal infection, which can increase and are neutral. However, it does not negatively affect mycorrhizal arbuscular fungal infections.
