Efek Preventif Pemberian Fortifikasi Ekstrak Rosella Ungu pada Yogurt Susu Sapi Terhadap Kadar MDA dan Gambaran Histopatologi Testis Tikus Putih (Rattus norvegicus) dengan Paparan Dioksin

Abstract

Dioksin merupakan zat toksik yang menimbukan dampak negatif terhadap organ reproduksi jantan. Fortifikasi bunga rosella ungu pada yogurt susu sapi memiliki efek antioksidan. Penelitian ini bertujuan untuk membuktikan potensi antioksidan dari yogurt rosella ungu pada paparan dioksin yang diamati melalui kadar MDA dan histopatologi organ testis. Hewan coba tikus Wistar (Rattus novergicus) jantan sebanyak 25 ekor dibagi menjadi lima kelompok: (1) kelompok kontrol negatif; (2) kelompok kontrol positif dengan pemberian TCDD; (3) kelompok perlakuan 1 diberikan TCDD dan yogurt susu sapi fortifikasi ekstrak bunga rosella ungu 0,5%; (4) kelompok perlakuan 2 diberikan TCDD dan yogurt susu sapi fortifikasi ekstrak bunga rosella ungu 1,0%; (5) kelompok perlakuan 3 diberikan TCDD dan yogurt susu sapi fortifikasi ekstrak bunga rosella ungu 1,5%. Semua perlakuan diberikan secara oral selama 12 hari. Pengujian kadar MDA diukur dengan metode Thiobarbituric Acid (TBA) dan pembuatan histopatologi menggunakan Hematoxylin eosin. Analisa data secara kuantitatif dengan one way ANOVA dan dilanjutkan dengan uji Tukey dengan taraf kepercayaan (p<0,05). Hasil riset membuktikan bahwa ekstrak rosella ungu 1% mengurangi pembentukan MDA di jaringan testis dan secara gambaran histopatologi semua konsentrasi belum mampu mencegah penurunan sel spermatogenik, sel Sertoli, dan sel Leydig. Kesimpulan dari penelitian ini adalah pemberian yogurt susu sapi fortifikasi ekstrak rosella ungu dapat mencegah kenaikan kadar MDA pada konsentrasi 1%, namun belum mampu mencegah penurunan setiap sel-sel di testis yang terpapar TCDD.

English Abstract

Dioxin is a toxic substance that has negative effects on male reproductive organs. Fortification of purple hibiscus flowers in yogurt has antioxidant effects. The study aims to prove the antioxidant potential of purple hibiscus yogurt in the presence of dioxin, as observed through MDA levels and testicular histopathology. The study involved 25 Wistar rats (Rattus novergicus) were divided into five groups: (1) negative control group; (2) positive control group given TCDD; (3) group 1 given TCDD and cow’s milk yogurt fortified with 0.5% purple hibiscus extract; (4) group 2 given TCDD and cow’s milk yogurt fortified with 1% purple hibiscus extract; (5) group 3 given TCDD and cow’s milk yogurt fortified with 1.5% purple hibiscus extract. All treatments were given orally for 12 days. MDA levels were measred using the Thiobarbituric Acid (TBA) method and histopathology was performed using Hematoxylin eosin. Quantitative analysis using one way ANOVA and followed by Tukey’s test with a confidence level of (p<0,05). The results of the study proved that 1% purple hibiscus extract reduces MDA formation in testicular tissue and in terms of histopathology, all concentrations were not able to prevent the decrease of spermatogenic cells, Sertoli cells, and Leydig cells. The conclusion of this study is that cow’s milk yogurt fortified with puple hibiscus extract can prevent an increase in MDA levels at a concentration of 1%, but is not yet able to prevent the decrease of all cells in the testes exposed to TCDD.
