Abstract

Pendahuluan: Kanker merupakan kondisi abnormal yang ditandai pertumbuhan sel ganas, tidak terkendali, dan mampu bermigrasi. Secara global, Insidensi kanker payudara pada tahun 2020 mencapai 2,26 juta jiwa. Tahapan migrasi sel kanker berkaitan erat dengan EMT pada tingkat seluler. Caulerpa racemosa adalah jenis anggur laut yang diyakini memiliki metabolit aktif bersifat antikanker dengan cara menghambat proliferasi dan menginduksi apoptosis. Tujuan: Mengetahui aktivitas antikanker ekstrak Caulerpa racemosa terhadap sel kanker payudara MCF-7 melalui mekanisme antimetastasis (uji migrasi sel dan ekspresi Snail). Metode: Studi eksperimental murni dengan rancangan post-test only controlled group secara in vitro. Subjek penelitian yaitu sel kanker payudara MCF-7 yang telah dikultur pada media DMEM, kemudian diujikan terhadap pemberian ekstrak Caulerpa racemosa (0, 100, 200, 400 μg/ml). Pengujian variabel penelitian dilakukan menggunakan metode scratch test dan pewarnaan imunofluoresensi. Data hasil pengujian dianalisis menggunakan SPSS dan Graphpad Prism 9. Hasil dan Pembahasan: Potensi penurunan laju migrasi dan ekspresi Snail pada sel kanker payudara MCF-7 paling besar ditemukan pada kelompok dosis 400 μg/ml. Hal ini dibuktikan dengan adanya jarak celah antar sel terlebar dan memiliki perbedaan signifikan (p < 0.05) dibandingkan semua kelompok eksperimen. Akan tetapi, pada penghambatan ekspresi Snail, perbedaan signifikan (p < 0.05) hanya ditemukan pada kelompok dosis 400 μg/ml terhadap kelompok kontrol. Kesimpulan: Pemberian ekstrak Caulerpa racemosa terbukti berpotensi sebagai agen antimetastasis sel kanker payudara MCF-7 melalui penurunan laju migrasi dan penghambatan ekspresi Snail. Studi lanjutan diperlukan untuk mengetahui aspek farmakologis dan zat aktif spesifik dari Caulerpa racemosa yang berperan sebagai antimetastasis.

English Abstract

Introduction: Cancer is an abnormal condition characterized by malignant, uncontrolled cell growth and the ability to migrate. Globally, the incidence of breast cancer in 2020 reached 2.26 million people. The stages of cancer cell migration are closely related to EMT at the cellular level. Caulerpa racemosa is a type of sea grape that is believed to have active anticancer metabolites by inhibiting proliferation and inducing apoptosis. Objective: To determine the anticancer activity of Caulerpa racemosa extract against MCF-7 breast cancer cells through antimetastatic mechanisms, specifically by conducting cell migration and Snail expression tests. Methods: The study was conducted in vitro using a true experimental design with a post-test-only controlled group. MCF-7 breast cancer cells were cultured in DMEM media and treated with different doses of Caulerpa racemosa extract (0, 100, 200, and 400 μg/mL). The scratch test and immunofluorescence staining were used to test the research variable, and the data collections were analyzed using SPSS and GraphPad Prism 9. Results and Discussion: The greatest potential reduction in migration rate and Snail expression in MCF-7 breast cancer cells was found in the 400 μg/mL dose group. The widest gap between cells proves this and has a significant difference (p < 0.05) compared to all experimental groups. However, in inhibiting Snail expression, a significant difference (p < 0.05) was only found in the 400 μg/mL dose group compared to the control group. Conclusion: The administration of Caulerpa racemosa extract was proven to have potential as an anti-metastatic agent for MCF-7 breast cancer cells by reducing migration rate and inhibiting Snail expression. Further studies are needed to determine the pharmacological aspects and specific active substances of Caulerpa racemosa, which act as antimetastases.
