Abstract

Kerusakan tanaman merupakan masalah yang sering disebabkan adanya serangan dari organisme pengganggu tanaman dari golongan hama dan patogen penyakit. Untuk melawan serangan OPT, tanaman mempunyai mekanisme pertahanan, termasuk adanya produksi metabolit sekunder. Metabolit sekunder tanaman merupakan hasil metabolit dari tanaman yang digunakan untuk perkembangbiakan maupun sebagai sistem pertahanan dari serangan hama dan patogen. Namun metabolit sekunder diproduksi oleh tanaman dengan terbatas dan tidak terus menerus. Salah satu cara untuk meningkatkan kandungan metabolit sekunder pada cabai yaitu dengan infeksi jamur antagonis dari golongan Trichoderma sp. sebagai stimulan. Bentuk infeksi ada berbagai cara, termasuk dengan perendaman benih menggunakan suspensi Trichoderma sp. Maka dari itu dilakukan analisis tentang kandungan senyawa metabolit sekunder tanaman cabai setelah infeksi jamur antagonis Trichoderma sp. dari hasil eksplorasi rizosfer dengan cara perendaman benih cabai dengan berbagai tingkatan lama waktu perendaman serta dikombinasikan dengan infeksi jamur patogen Fusarium sp. Kegiatan penelitian dilaksanakan pada bulan Januari hingga Maret 2023 di Laboratorium Penyakit Tumbuhan 3 dan Laboratorium Toksikologi, Fakultas Pertanian, Universitas Brawijaya. Penelitian dilakukan melalui beberapa tahapan yaitu persiapan penelitian dengan pembuatan media PDA, peremajaan jamur antagonis dan patogen, pembuatan PDB, pembuatan suspensi jamur, perendaman benih cabai dengan suspensi Trichoderma sp., perendaman benih cabai dengan suspensi Fusarium sp., dan perendaman benih cabai dengan suspensi Trichoderma sp. dan Fusarium sp. di dua hari berikutnya. Perendaman menggunakan perlakuan lama perendaman yaitu 10, 20, dan 30 menit. Lalu dilakukan proses perkecambahan selama 14 hari dan diakhiri dengan uji fitokimia Harbone (1996) yang terdiri dari pengujian alkaloid, flavonoid, tanin, saponin, steroid, dan triterpenoid.

English Abstract

Plant damage is a problem that is often caused by attacks from plantdisturbing organism class of pests and disease pathogens. To fight pest attacks, plants have defense mechanisms, including the production of secondary metabolites. Plant secondary metabolites are the results of metabolites from plants that are used for breeding and as a defense system against pests and pathogens. However, secondary metabolites are produced by plants limitedly and not continuously. One way to increase the content of secondary metabolites in chili is by infection with antagonistic fungi from the Trichoderma sp. as a stimulant. There are various forms of infection, including by soaking the seeds using Trichoderma sp. suspension. Therefore, an analysis was carried out on the content of secondary metabolites of chili plants after infection with the antagonistic fungus Trichoderma sp. from the results of exploration of the rhizosphere by soaking chili seeds with various levels of soaking time and combined with infection with the pathogenic fungus Fusarium sp. Research activities were carried out from January to March 2023 at the Laboratory of Plant Diseases 3 and the Laboratory of Toxicology, Faculty of Agriculture, Brawijaya University. The research was conducted through several stages, namely research preparation by making PDA media, antagonistic and pathogenic fungal rejuvenation, PDB preparation, fungus suspension preparation, chili seed soaking with Trichoderma sp. suspension, chili seed soaking with Fusarium sp. suspension, and chili seed soaking with suspension Trichoderma sp. and Fusarium sp. the next two days. Soaking using long immersion treatment that is 10, 20, and 30 minutes. Then the germination process was carried out for 14 days and ended with Harbone's (1996) phytochemical test which consisted of testing alkaloids, flavonoids, tannins, saponins, steroids, and triterpenoids.
