Abstract
Virus panleukopenia (Feline panleukopenia virus) adalah sebuah spesies dari virus parvo yang dapat menginfeksi semua kucing, baik kucing liar maupun kucing peliharaan. Virus ini tidak beramplop, genom DNA untai tunggal dan memiliki dua protein struktural, yaitu: VP1 dan VP2 dan dua protein non struktural, yaitu: NS1 dan NS2. Penelitian ini bertujuan untuk memperoleh informasi ilmiah dari karakteristik FPV isolat lokal terbaru dan menjelaskan homologi genetik FPV isolat lokal dibandingan dengan isolat vaksin strain Johnson Snow Leopard yang digunakan di Indonesia berdasarkan gen VP1 dan VP2. Sekuen ssDNA gen VP1 dan VP2 diperoleh dari satu sampel vaksin dan lima sampel swab rektum kucing yang terinfeksi FPV dilakukan tahap selanjutnya yaitu tahap isolasi DNA dan amplifikasi dengan metode PCR menggunakan sepasang primer forward 5’- CAG GAA GAT ATC CAG AAG GA -3’ dan reverse 5’- GGT GCT AGT TGA TAT GTA ATA AAC A -3’. Hasil dari sekuens perolehan dari amplifikasi dianalisa untuk memperlihatkan hubungan kekerabatan antar sampel, serta membandingkan gen VP1 dan VP2 pada isolat lapang dan isolat vaksin strain Johnson Snow Leopard menggunakan program BioEdit®, Basic Local Aligment Search Tool (BLAST) dari NCBI, dan program Molecular Evolutionary Genetics Analysis (MEGA). Hasil alignment sekuens sampel FPV isolat vaksin dengan sampel FPV isolat lapang terdapat mutasi di beberapa lokasi. Jarak genetik sampel FPV isolat vaksin dibandingkan dengan isolat lapang bernilai ≤ 0,14%. Kesimpulan dari penelitian ini adalah berdasarkan hasil sekuen DNA memiliki hubungan kekerabatan antara sampel FPV isolat vaksin (T1) dengan sampel FPV isolat lapang (T2, T3, T4, T5 dan T6) yang berada di daerah Jawa Timur.
English Abstract
Panleukopenia virus (Feline panleukopenia virus) is a species of parvovirus that can infect all wild and domestic cats. This virus has non-enveloped protein capsids, genomes are single stranded DNA, has two structural proteins i.e. VP1 and VP2, and has two nonstructural proteins i.e. NS1 and NS2. The purpose of this research is to give scientific information about the characteristic of FPV latest local isolate and to explain FPV genetic homology local isolate compared with FPV vaccine isolate which has been used in Indonesia based on ssDNA sequences of VP1 and VP2 genes. The ssDNA sequences of VP1 and VP2 was obtained from one sample of FPV vaccine isolate and five swab rectum samples of cats that has been suspected with FPV after passing through the DNA isolation and amplification stages using PCR with forward primer 5'- CAG GAA GAT ATC CAG AAG GA -3 ' and reverse 5'- GGT GCT AGT TGA TAT GTA ATA AAC A -3 '. Sequences obtained from ssDNA sequence amplification are analyzed to see the relationship then compared with ssDNA sequence from vaccine isolate strain Johnson Snow Leopard and local isolates using BioEdit®, Basic Search Aligment Search Tool (BLAST) from the NCBI, and the Molecular Evolutionary Genetics Analysis (MEGA) program. The results of sequences alignment with BioEdit of FPV vaccine isolate with field isolates showed mutations in several locations. The genetic distances of FPV in vaccine isolate compared with local isolates less than or equal to ≤ 0,14%. The conclusion of this study is based on the results of DNA sequences, there is a relationship between the vaccine isolate (T1) and the field isolate samples (T2, T3, T4, T5 and T6) located in East Java.
