Abstract
Streptococcus mutans merupakan bakteri yang terdapat dalam rongga mulut yang berperan sebagai agen utama penyebab karies gigi, yang menghasilkan enzim glukosiltransferase (GTF). Enzim GTF mengubah sukrosa menjadi fruktosa dan glukosa. Bunga telang (Clitoria ternatea) sebagai bahan alami memiliki sifat antibakteri. Dalam hal ini memiliki kandungan flavonoid dan fenolik yang dapat menghambat aktivitas enzim GTF Streptococcus mutans. Tujuan penelitian ini untuk mengetahui pengaruh air seduhan bunga telang (Clitoria ternatea) terhadap aktivitas enzim glukosiltransferase (GTF) Streptococcus mutans. Penelitian ini menggunakan seduhan bunga telang (Clitoria ternatea) konsentrasi 50%, 75%, dan 100% sebagai perlakuan, chlorhexidine gluconate 0,12% sebagai kontrol positif dan akuades steril sebagai kontrol negatif. Enzim GTF diperoleh dari hasil sentrifugasi bakteri Streptococcus mutans. Aktivitas enzim GTF diukur dengan menghitung kadar fruktosa menggunakan High Performance Liquid Chromatography (HPLC). Hasil perhitungan aktivitas enzim GTF menunjukkan perbedaan penurunan kadar fruktosa yang signifikan antara kelompok perlakuan yang diberikan seduhan bunga telang dengan kelompok kontrol. Analisa data menggunakan korelasi dan regresi menunjukkan pengaruh sebesar 98% terhadap penurunan kadar fruktosa. Berdasarkan penelitian bunga telang dapat menghambat kerja dari enzim glukosiltransferase.
English Abstract
Streptococcus mutans is a bacteria in the oral cavity which consist of glucosyltransferase (GTF) enzyme and acts as the main agent that cause dental caries. GTF enzyme will convert sucrose into fructose and glucose. Butterfly pea (Clitoria ternatea) flower is one of herbal plants which has a antibacterial effect. Butterfly pea flower has active substance such as flavonoids and phenolic which specifically can inhibit GTF enzyme Streptococcus mutans. The purpose of this research is to investigate the effect of steeping of the flower of butterfly pea (Clitoria ternatea) to the activity of Streptococcus mutans’s glucosyltransferase (GTF) enzyme. This research used 50%, 75%, and 100% concentration of steeping of butterfly pea flower as the treatment groups, 0,12% chlorhexidine gluconate as a positive control, and distillate water as a negative control. The GTF enzyme is obtained from supernatant of Streptococcus mutans. The activity of GTF enzyme is measured by calculating the fructose concentration using High Performance Liquid Chromatography (HPLC). The results of calculations of enzyme activity GTF showed a significant difference of fructose levels between treatment groups given the steeping of butterfly pea flower with the control groups. Data analysis using correlation and regression shows the effect of 98% againts a decrease in levels of fructose. Based on this research, it can beconclude that steeping of butterfly pea flower can inhibit the action of the glucosiltransferase enzyme.
