Abstract
Padi (Oryza sativa L.) merupakan tanaman budidaya yang mayoritas ditanam di berbagai wilayah Indonesia. Ketergantungan dan konsumsi beras pada masyarakat sangat tinggi, sedangkan dalam kurun waktu 2015-2019 pertumbuhan produksi padi mengalami penurunan rata-rata sebesar 4,18% per tahun. Salah satu faktornya yaitu serangan penyakit Hawar Daun Bakteri (HDB) yang disebabkan oleh Xanthomonas oryzae pv. oryzae. dan menyebabkan kerusakan sebesar 50%. Penggunaan pestisida yang tidak bijaksana dan tidak berdasarkan pemantauan berdampak buruk terhadap lingkungan. Oleh karena itu perlu dilakukan pengendalian penyakit tanaman padi yang ramah lingkungan. Tujuan penelitian ini yaitu mendapatkan bakteri filoplan potensial antagonis terhadap patogen X. oryzae, mengetahui potensi daya hambat terhadap patogen X. oryzae dan mengetahui karakteristik isolat bakteri filoplan antagonis. Penelitian dilaksanakan di Laboratorium Penyakit Tumbuhan 1, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya Malang pada bulan Maret 2020 sampai dengan Maret 2021. Metode yang digunakan antara lain: pengambilan sampel; eksplorasi bakteri filoplan; peremajaan bakteri X. oryzae; uji patogenesitas; seleksi antagonis; uji hipersensitif; uji antagonis; identifikasi dan karakterisasi. Variabel pengamatan terdiri atas morfologi dan zona hambat bakteri filoplan antagonis. Terdapat 80 isolat bakteri hasil eksplorasi dari daun bendera tanaman padi organik. Seleksi bakteri filoplan potensial antagonis terhadap X. oryzae berjumlah 27 isolat bakteri dan dipilih 5 isolat bakteri dengan zona penghambatan terbesar. Hasil pengujian antagonis menunjukan bahwa isolat bakteri dengan kode LS2, LS5, LS6, LS7 dan LS40 mempunyai kemampuan lebih baik dari bakterisida berbahan aktif Streptomycin sulfate 20%. Hasil identifikasi dan karakterisasi menunjukan kelima isolat bakteri termasuk kedalam genus Erwinia sp.
English Abstract
Rice (Oryza sativa L.) is a cultivated plant that is predominantly grown in various regions of Indonesia. The dependency and consumption of rice in society are very high, while within the period of 2015-2019, rice production experienced an average decline of 4,18% per year. One of the contributing factors is the attack of Bacterial Leaf Blight Disease (BLB) caused by Xanthomonas oryzae pv. oryzae, which results in a 50% damage rate. The majority of farmers use synthetic chemical pesticides to control BLB disease. The indiscriminate and unmonitored use of pesticides has detrimental effects on the environment. The purpose of this research is to obtain potential phylloplane bacteria that act as antagonists against the X. oryzae pathogen, to determine the inhibitory potential against the X. oryzae pathogen, and to understand the characteristics of antagonistic phylloplane bacterial isolates. The research was conducted at Plant Disease Laboratory 1, Department of Plant Pests and Diseases, Faculty of Agriculture, Brawijaya University Malang from March 2020 to March 2021. The methods used included: sample collection; exploration of phylloplane bacteria; rejuvenation of X. oryzae bacteria; pathogenicity tests; antagonist selection; hypersensitive tests; antagonist tests; identification and characterization. The observation variables consisted of morphology and inhibition zones of antagonistic phylloplane bacteria. There were 80 bacterial isolates resulting from the exploration of organic rice plant flag leaves. The selection of potential phylloplane bacteria antagonists against X. oryzae yielded 27 bacterial isolates, and 5 bacterial isolates were chosen based on the largest inhibition zones. The results of the antagonist testing showed that isolates with codes LS2, LS5, LS6, LS7 and LS40 had better abilities than the bactericide with the active ingredient Streptomycin sulfate 20%. The results of identification and characterization indicated that all five bacterial isolates belong to the genus Erwinia sp.
