Abstract
Penyakit ikan dapat mengganggu atau menghambat kegiatan budidaya Adanya penyakit ikan di kolam terjadi karena ketidakseimbangan antara lingkungan, ikan dan patogen. Penyakit yang sering ditemukan pada budidaya adalah penyakit yang disebabkan oleh bakteri. Salah satu contoh bakteri yang sering ditemukan yakni bakteri Vibrio alginolyticus menyerang ikan kerapu dan udang. Permasalahan penyakit yang disebabkan bakteri dapat diatasi dengan manajemen kesehatan ikan melalui usaha pengendalian penyebaran infeksi. Pengendalian yang biasa dilakukan yaitu dengan pemberian antibakteri seperti bahan-bahan antibiotik melalui kegiatan pengobatan. Pemberian antibiotik secara terus menerus dapat mengakibatkan resistensi bakteri terhadap antibiotik dan mencemari hasil budidaya. Alternatif pengobatan dari bahan alami seperti daun kemangi mengandung antibakteri seperti flavonoid, tannin, saponin yang bisa larut dalam air dapat merusak membran sitoplasma dan mendenaturasi protein sel. Penelitian dilakukan di Laboratorium Budidaya Ikan Divisi Penyakit dan Kesehatan Ikan, Fakultas Perikanan dan Ilmu Kelautan, Universitas Brawijaya, Malang. Penelitian dilaksanakan pada bulan Desember 2022 - Januari 2023. Tujuan penelitian ini adalah untuk mengetahui daya hambat ekstrak kasar daun kemangi (O. sanctum) terhadap bakteri V. alginolyticus in vitro. Metode penelitian yang digunakan yaitu metode eksperimental. Metode yang meguji secara langsung terhadap pengaruh variabel terhadap variabel lain dan menguji hipotesis dengan hubungan sebab akibat. Rancangan percobaan yang digunakan adalah RAL dengan menggunakan 5 perlakuan dan 3 ulangan. Dosis perlakuan ekstrak kasar daun kemangi (O. sanctum) yaitu: A (15 ppm), B (30 ppm), C (45 ppm), D (60 ppm) dan E (75 ppm). Hasil penelitian menunjukan bahwa ekstrak kasar daun kemangi (O. sanctum) memberikan pengaruh berbeda nyata terhadap daya hambat pertumbuhan bakteri V. alginolyticus dalam uji cakram. Perlakuan A (15 ppm) dengan rerata zona hambat 7,34 ± 0,37 mm menjadi perlakuan dengan rerata zona hambat terkecil dan dosis terbaik pada penelitian ini yaitu perlakuan D (60 ppm) dengan rerata zona hambat 8,32 ± 0,07 mm. Hubungan antara dosis ekstrak kasar daun kemangi dengan diameter zona hambat yang terbentuk adalah grafik linier dimana persamaan y= 0,0214x + 6,939 dengan koefisien determinasi (R2) sebesar 0,6462. Kesimpulan hasil penelitian adalah ekstrak kasar daun kemangi (O. sanctum) dapat menghambat bakteri V. alginolyticus secara in vitro pada dosis terbaik sebesar 60 ppm. Ekstrak kasar daun kemangi (O. sanctum) bersifat bakterisidal terhadap bakteri V. alginolyticus. Diameter zona hambat ekstrak kasar daun kemangi berkisar antara 7,34 ± 0,37 mm – 8,64 ± 0,33 mm yang berarti ekstrak kasar daun kemangi (O. sanctum) termasuk kategori aktivitas antibakteri sedang.
English Abstract
Fish diseases can interfere with or hinder aquaculture activities. The presence of fish diseases in ponds occurs due to an imbalance between the environment, fish and pathogens. Diseases that are often found in cultivation are diseases caused by bacteria. One example of bacteria that is often found is Vibrio alginolyticus attacking grouper and shrimp. Disease problems caused by bacteria can be overcome by fish health management through efforts to control the spread of infection. The usual control is by administering antibacterial such as antibiotic ingredients through treatment activities. Continuous administration of antibiotics can result in bacterial resistance to antibiotics and contaminate cultivated products. Alternative treatment from natural ingredients such as basil contains antibacterial such as flavonoids, tannins, saponins that can dissolve in water can damage the cytoplasmic membrane and denature cell proteins. The research was conducted at Laboratorium Budidaya Ikan Divisi Penyakit dan Kesehatan Ikan, Fakultas Perikanan dan Ilmu Kelautan, Universitas Brawijaya, Malang. The research was conducted in December 2022 - January 2023. This study aims to determine the inhibitory of basil’s crude leaf (O. sanctum) extract against V. alginolyticus bacteri by in vitro. The research method used is the experimental method. Amethod that directly examines the influence of variables on other variables and tests hypotheses with causal relationships. The experimental design used was RAL using 5 treatments and 3 repeats. The treatment dose of crude basil leaf extract (O. sanctum) is: A (15 ppm), B (30 ppm), C (45 ppm), D (60 ppm) and E (75 ppm). The results showed that crude extract of basil leaves (O. sanctum) had a significantly different effect on the growth inhibition of V. alginolyticus bacteria in disc tests. Treatment A (15 ppm) with an average inhibitory zone of 7.34 ± 0.37 mm became the treatment with the smallest average inhibitory zone and the best dose in this study was treatment D (60 ppm) with an average inhibitory zone of 8.32 ± 0.07 mm. The relationship between the dose of crude basil leaf extract and the diameter of the inhibitory zone formed is a linear graph where the equation y = 0.0214x + 6.939 with a coefficient of determination (R2) of 0.6462. The conclusion of the results of the study is that crude extract of basil leaves (O. sanctum) can inhibit V. alginolyticus by in vitro at the best dose of 60 ppm. Crude extract of basil leaves (O. sanctum) is bactericidal against V. alginolyticus. The diameter of the inhibitory zone of crude basil leaf extract ranges from 7.34 ± 0.37 mm – 8.64 ± 0.33 mm which means that crude basil leaf extract (O. sanctum) belongs to the category of medium antibacterial activity.
