Abstract

Penggunaan kontrasepsi hormonal terkadang menimbulkan efek samping, sehingga menjadi pertimbangan bagi wanita untuk menggunakan kontrasepsi hormonal. Sehingga dapat dicari alternatif lain yang memiliki efek samping minimal. Hal inilah yang menjadi latar belakang penelitian menggunakan tanaman sebagai antifertilitas yang aman dengan efek yang minimal. Daun tanaman ruta (Ruta angustifolia L. Pers) mengandung senyawa yang diduga mampun mempengaruhi kerja hormonal sehingga dipertimbangkan sebaga senyawa antifertilitas seperti tanin, saponin dan flavonoid. Penelitian ini untuk menguji pengaruh ekstrak etanol daun ruta terhadap folikel de graaf, folikel atresia dan toksisitasnya terhadap hepar dan ginjal. Penelitian ini menggunakan bahan biologis yang disimpan dan pengamatan histopatologi pada bulan November 2022. Tikus strain Wistar betina digunakan sebagai hewan coba serta menggunakan 4 kelompok yakni (tanpa perlakuan; dosis 0,4; 0,8; 1,2 mg/g BB). Folikel de graaf, folikel atresia dan nekrosis hati dan ginjal diukur pada slide histopatologis dengan pewarnaan HE menggunakan mikroskop tipe Olympus CX23 dan aplikasi ImageJ. Pada P2 jumlah folikel de graaf turun namun tidak signifikan sedangkan jumlah folikel atresia meningkat signifikan (p=0,014). Penggunaan daun ruta tergolong aman pada semua dosis untuk hepar. Jumlah nekrosis sel ginjal terendah ditemukan pada P1 dengan hasil yang tidak signifikan. Penurunan jumlah folikel de graaf dan peningkatan jumlah folikel atresia disebabkan pemberian ekstrak etanol daun rue dengan dosis optimal 0,8 g/kg BB. Jumlah nekrosis sel ginjal menurun akibat pemberian ekstrak etanol daun ruta dengan dosis optimal 0,4 g/kg BB. Di hati, penggunaan ruta aman untuk semua dosis.

English Abstract

The use of hormonal contraception sometimes causes side effects, this is consideration for women to use hormonal contraception. So that other alternatives can be sought which have minimal side effects. This is the background for research using plants as antifertility agents that are safe with minimal effects. The leaves of the ruta plant (Ruta angustifolia L. Pers) contain compounds that are thought to be able to influence hormonal action so that they are considered as antifertility compounds such as tannins, saponins and flavonoids. This study was to examine the effect of ruta leaf ethanol extract on de graaf follicles, atretic follicles and its toxicity to the liver and kidneys. This study used stored biological material and histopathological observations in November 2022. Wistar female rats were used as experimental animals and used 4 groups namely (no treatment; doses 0.4; 0.8; 1.2 mg/g BW). Graafian follicles, atretic follicles and hepatic and renal necrosis were measured on histopathological slides by HE staining using an Olympus CX23 type microscope and ImageJ application. At P2 the number of de graaf follicles decreased but not significantly while the number of atretic follicles increased significantly (p=0.014). The use of ruta leaves is classified as safe at all doses for the liver. The lowest amount of kidney cell necrosis was found in P1 with insignificant results. The decrease in the number of de graaf follicles and the increase in the number of atretic follicles was due to administration of ethanol extract of rue leaves at an optimal dose of 0.8 g/kg BW. The amount of kidney cell necrosis decreased due to the administration of ethanol extract of rue leaves at an optimal dose of 0.4 g/kg BW. In the liver, use of rue is safe for all doses.
