Abstract

Penyakit malaria memiliki prevalensi tinggi di Indonesia, khususnya bagian timur. Salah satu tanaman herbal yang digunakan sebagai antimalaria adalah tanaman songga (Strychnos lucida R.Br.). Tujuan penelitian ini untuk mengetahui aktivitas antimalaria ekstrak etil asetat batang songga terhadap Plasmodium berghei ANKA secara in vivo. Songga dimaserasi dengan pelarut etil asetat lalu dipreparasi menjadi tiga dosis yaitu 1, 10, 100 mg/kgBB. Setelah itu dilakukan uji aktivitas antimalaria secara in vivo. Metode yang digunakan yaitu Peter’s test, hewan coba diberikan larutan uji selama 4 hari berturut-turut dan dibuat hapusan darah melalui ekor mencit selama 7 hari. Terdapat tiga parameter yang diamati yaitu, persen parasitemia, persen pertumbuhan parasitemia, dan persen penghambatan parasitemia. Hasil penelitian menunjukkan rata-rata persen parasitemia hari ke-6 pada perlakuan ekstrak dosis 1, 10, 100 mg/kgBB berturutturut 15,77%, 13,82%, dan 13,16%. Sedangkan rata-rata persen pertumbuhan parasitemia pada perlakuan ekstrak dosis 1, 10, 100 mg/kgBB berturut-turut 2,50%, 2,12%, dan 2,08%. Rata-rata persen penghambatan parasitemia pada perlakuan ekstrak dosis 1, 10, 100 mg/kgBB berturut-turut 29,23%, 37,96%, 40,49%. Berdasarkan uji Spearman yang dilakukan terdapat korelasi yang bermakna antara dosis ekstrak yang diberikan terhadap persen parasitemia, persen pertumbuhan, dan persen penghambatan. Nilai r berturut-turut -0,395, - 0,603, dan 0,625. Semakin tinggi dosis ekstrak yang diberikan, maka semakin rendah persen parasitemia dan persen pertumbuhannya, namun semakin tinggi persen penghambatannya. Berdasarkan uji Kruskall Wallis terdapat beberapa hubungan yang tidak signifikan antar kelompok perlakuan ekstrak maupun kontrol positif dan negatif. Sehingga dapat dilihat bahwa pemberian ekstrak etil asetat batang songga menunjukkan adanya kemampuan menghambat pertumbuhan parasit namun tidak signifikan.

English Abstract

Malaria has a high prevalence in Indonesia, especially in the eastern part. One of the herbal plants used as antimalarial is songga (Strychnos lucida R.Br.). The aim of this study is to determine the antimalarial activity of ethyl acetate extract of songga stems against Plasmodium berghei ANKA by in vivo. Songga was macerated with ethyl acetate solvent and then prepared into three doses, namely 100, 10, 1 mg/kgBW. After that, an in vivo antimalarial activity test was carried out. The method used was the Peter's test, the experimental animals were given a test solution for 4 consecutive days and blood smears were made through the tails of mice for 7 days. There are three parameters observed, namely, the percentage of parasitemia, the percentage of parasitemia growth, and the percentage of parasitemia inhibition. The results showed that the average percentage of parasitemia on the 6th day at extract treatment at doses of 1, 10, 100 mg/kgBW were 15.77%, 13.82% and 13.16%, respectively. Meanwhile, the average percentage growth of parasitemia in extract treatment at doses of 1, 10, 100 mg/kg BW were 2.50%, 2.12%, and 2.08%, respectively. The average percentage of inhibition of parasitemia in extract treatment at doses of 1, 10, 100 mg/kgBW were 29.23%, 37.96%, 40.49%, respectively. Based on the Spearman test conducted, there was a significant correlation between the dose of the extract given to the percentage of parasitemia, the percentage of growth, and the percentage of inhibition. The r values are -0.395, -0.603, and 0.625 respectively. The higher the dose of the extract given, means the lower the percentage of parasitemia and the percentage of growth, but the higher the percentage of inhibition. Based on the Kruskall Wallis test, there were several insignificant relationships between the extract treatment groups and the positive and negative controls. So it can be be concluded that the administration of ethyl acetate extract of Songga stems showed the ability to inhibit parasite growth but not significantly.
