Abstract

Latar belakang: Preeklamsia adalah penyakit ibu hamil yang terjadi diatas usia kehamilan 20 minggu, diketahui dengan adanya tekanan darah sistolik ≥ 140 mmHg sedangkan diastolik > 90 mmHg, dengan atau tanpa proteinuria dalam waktu 24 jam melebihi 0,3 gr/L. Preeklamsia dapat menyebabkan kematian pada ibu dan janin. Disfungsi endotel akibat stress oksidatif menyebabkan penurunan kadar NO dalam darah, akibatnya muncul hipertensi sebagai manifestasi klinis preeklamsia. Kurkumin sebagai antioksidan dapat mengurangi radikal bebas dan memperbaiki fungsi endotel. Formulasi nano kurkumin dapat meningkatkan aktivitas farmakologis kurkumin. Tujuan: Mengetahui pengaruh pemberian nano kurkumin terhadap tekanan darah pada tikus putih strain wistar (Rattus norvegicus) bunting model preeklamsia. Metode: True experimental dengan menggunakan desain penelitian post-test only control group. Sampel yang digunakan sebanyak 24 tikus bunting yang terbagi kedalam kelompok kontrol negatif dan positif serta 4 kelompok perlakuan dengan pemberian 4 dosis nano kurkumin berbeda. Pemberian L-NAME dan nano kurkumin dilakukan mulai usia kehamilan hari ke 13-18. Data tekanan darah diambil saat usia kehamilan 19 hari. Hasil: Terdapat perbedaan yang signifikan pada tekanan darah masing�masing kelompok perlakuan berdasarkan Uji One-Way ANOVA (p-value = 0,000). Pada uji LSD terlihat bahwa kelompok P1, P2, dan P3 berpengaruh signifikan (p-value <0,05). Kelompok P1 adalah kelompok dengan dosis nano kurkumin paling efektif untuk mencegah peningkatan tekanan darah. Kesimpulan: Pemberian nano kurkumin dosis 25 mg, 50 mg, dan 100 mg dapat mencegah peningkatan tekanan darah pada tikus bunting dengan kondisi preeklamsia. Nano kurkumin 25 mg adalah dosis yang paling efektif.

English Abstract

Background: Preeclampsia is a disease of pregnant women that occurs above 20 weeks of gestation, known by the presence of systolic blood pressure ≥ 140 mmHg while diastolic > 90 mmHg, with or without proteinuria within 24 hours exceeding 0.3 gr/L. Preeclampsia can cause death to the mother and fetus. Endothelial dysfunction due to oxidative stress causes a decrease in NO levels in the blood, resulting in hypertension as a clinical manifestation of preeclampsia. Curcumin as an antioxidant can reduce free radicals and improve endothelial function. Nano curcumin formulation can increase the pharmacological activity of curcumin. Objective: To determine the effect of nano curcumin administration on blood pressure in pregnant white rat wistar strain (Rattus norvegicus) model of preeclampsia. Method: True experimental, by using a post-test only control group research design. There were 24 pregnant rats included in the negative and positive control groups and 4 treatment groups with 4 different doses of nano curcumin. L-NAME and nano curcumin is given from the 13th day to the 18th day of gestation, while the data of blood pressure was collected at 19 days of gestation. Result: There was a significant difference in the blood pressure of each treatment group based on the One-Way ANOVA test (p-value = 0.000). The LSD test showed that group of P1, P2, and P3 have significant effect (p-value <0.05). The P1 group was the group with the most effective dose of nano�curcumin to prevent an increase in blood pressure. Conclusion: Administration of nano-curcumin at doses of 25 mg, 50 mg, and 100 mg can prevent an increase in blood pressure in pregnant rat with preeclampsia. 25 mg of nano curcumin is the most effective dose
