Abstract

Ikan kerapu hibrida cantang (Epinephelus sp.) merupakan salah satu komoditas perikanan yang memiliki nilai ekonomis tinggi. Ikan kerapu cantang merupakan hasil perkawinan silang antara ikan kerapu macan (Epinephelus fuscogutattus) betina dengan ikan kerapu kertang (Epinephelus lanceoulatus) jantan. Virus Viral Nervous Necrosis merupakan salah satu virus yang menyerang komoditas perikanan laut. Virus Viral Nervous Necrosis pertama kali menyerang komoditas perikanan di Indonesia pada tahun 1997 dan terus menyebar hingga saat ini. Salah satu upaya pencegahan infeksi virus Viral Nervous Necrosis adalah dengan memberikan nanovaksin rekombinan Chlorella vulgaris berbasis silver nanopartikel C-AgNPs. Tujuan dari penelitian ini adalah mengetahui pengaruh pemberian nanovaksin rekombinan Chlorella vulgaris berbasis silver nanopartikel C-AgNPs terhadap profil sel darah dan pertumbuhan ikan kerapu cantang (Epinephelus sp.) yang terinfeksi virus Viral Nervous Necrosis. Metode yang digunakan dalam penelitian ini adalah metode eksperimen Rancangan Acak Lengkap (RAL) faktorial 8 perlakuan dan 3 kali ulangan yaitu (K+) ikan kerapu cantang + VNN ; (K-) Ikan kerapu cantang sehat ; (A1B1) Ikan kerapu cantang + nanovaksin 33 μl ; (A1B2) Ikan kerapu cantang + nanovaksin 66 μl ; (A1B3) Ikan kerapu cantang + nanovaksin 112 μl ; (A2B1) Ikan kerapu cantang + nanovaksin 33 μl + virus VNN ; (A2B2) Ikan kerapu cantang + nanovaksin 66 μl + virus VNN ; (A2B3) Ikan kerapu cantang + nanovaksin 112 μl + virus VNN yang diamati selama 28 hari. Penelitian dilaksanakan pada bulan November 2022 hingga bulan Januari 2023 di Laboratorium Demoplot Pengujian Nanovaksin Riset Kolaborasi Indonesia. Selama masa penelitian, dilakukan pengambilan dan pengamatan sampel profil darah ikan yaitu sel eritrosit, sel leukosit, kadar hemoglobin, diferensial leukosit (limfosit, monosit, basofil dan neutrofil),dan juga dilakukan pengamatan pertumbuhan sesaat, pertumbuhan relatif, dan pertumbuhan total selama 28 hari. Analisis data yang digunakan dalam penelitian ini adalah Analysis of Variance (ANOVA) yang dilanjutkan dengan Uji Least Significant Difference (LSD) dan Uji Duncan. Hasil peneltian menunjukkan bahwa pemberian nanovaksin rekombinan Chlorella vulgaris berbasis silver nanopartikel C-AgNPs memberikan pengaruh yang cukup signifikan dalam meningkatkan gen imunitas pasca uji tantang pada ikan kerapu cantang (Epinephelus sp.) selama 28 hari dilihat dari respon seluler darah ikan. Pada perlakuan A2B1 (Ikan kerapu cantang + nanovaksin 33 μl + virus VNN) memberikan hasil penelitian terbaik dengan pertumbuhan sesaat (berat: 0,022 ; panjang: 0,014), pertumbuhan relatif (berat: 83,3 % ; panjang: 50 %), pertumbuhan total (berat: 20% ; panjang: 6%). Jumlah sel eritrosit terbaik didapatkan pada A2B1 (2,55x106 sel/mm3). Jumlah sel leukosit terbaik didapatkan pada A2B1 (1,80x104 sel/mm3). Kadar Hb terbaik didapatkan pada A2B1 (6,7 g/dL). Jumlah sel diferensial leukosit terbaik didapatkan pada A2B1 yaitu (limfosit: 89% ; monosit: 23,33% ; basofil: 0,10% ; neutrofil: 22%). Kesimpulan dari penelitian ini adalah pemberian nanovaksin rekombinan Chlorella vulgaris berbasis silver nanopartikel C-AgNPs dapat digunakan dalam pencegahan infeksi virus Viral Nervous Necrosis dan dosis terbaik pemberian nanovaksin adalah 33 μl yang merupakan level imunogenitas vaksin yang efektif dalam meningkatkan sistem imunitas tubuh ikan kerapu cantang (Epinephelus sp.) dilihat dari respon seluler darah ikan kerapu cantang (Epinephelus sp.). Diperlukan penelitian lebih lanjut mengenai pengaruh nanovaksin rekombinan berbasis nanopartikel CAgNPs secara langsung pada ikan kerapu cantang di keramba jaring apung atau kolam budidaya untuk mengendalikan pathogen melalui peningkatan sistem imunitas ikan yaitu sel darah putih (leukosit) dan diferensial leukosit.

English Abstract

The cantang hybrid grouper (Epinephelus sp.) is a fishery commodity that has high economic value. The cantang grouper is the result of crossbreeding between female tiger grouper (Epinephelus fuscogutattus) and male kertang grouper (Epinephelus lanceoulatus). Viral Nervous Necrosis is one of the viruses that attack marine fisheries commodities. Viral Nervous Necrosis first attacked fishery commodities in Indonesia in 1997 and continues to spread until now. One of the efforts to prevent Viral Nervous Necrosis infection is by giving recombinant Chlorella vulgaris nanovaccines based on silver nanoparticles C-AgNPs. The purpose of this study was to determine the effect of giving recombinant Chlorella vulgaris nanovaccines based on silver nanoparticles C-AgNPs on blood cell profiles and growth of grouper cantang (Epinephelus sp.) infected with Viral Nervous Necrosis. The method used in this study is an experimental method of Completely Randomized Design (CRD) factorial 8 treatments and 3 replicates, namely (K +) hybrid grouper + VNN; (K-) healthy grouper; (A1B1) hybrid grouper + 33 μl nanovaccine; (A1B2) hybrid grouper + 66 μl nanovaccine; (A1B3) hybrid grouper + 112 μl nanovaccine; (A2B1) hybrid grouper + 33 μl nanovaccine + VNN virus; (A2B2) hybrid grouper + 66 μl nanovaccine + VNN virus; (A2B3) hybrid grouper + 112 μl nanovaccine + VNN virus observed for 28 days. The study was conducted from November 2022 to January 2023 at the Indonesian Collaborative Research Nanovaccine Testing Demoplot Laboratory. During the study period, fish blood profile samples were taken and observed, namely erythrocyte cells, leukocyte cells, hemoglobin levels, leukocyte differential (lymphocytes, monocytes, basophils and neutrophils), and also observed instantaneous growth, relative growth, and total growth for 28 days. Data analysis used in this study was Analysis of Variance (ANOVA) followed by Least Significant Difference (LSD) test and Duncan test. The results showed that the administration of recombinant Chlorella vulgaris nanovaccines based on silver nanoparticles C-AgNPs had a significant effect in increasing post-challenge immunity genes in cantang grouper (Epinephelus sp.) for 28 days seen from the cellular response of fish blood. The A2B1 treatment (hybrid grouper + 33 μl nanovaccine + VNN virus) gave the best research results with instantaneous growth (weight: 0.022; length: 0.014), relative growth (weight: 83.3%; length: 50%), total growth (weight: 20%; length: 6%). The best erythrocyte cell count was obtained at A2B1 (2.55x106 cells/mm3). The best leukocyte cell count was obtained at A2B1 (1.80x104 cells/mm3). The best Hb level was obtained at A2B1 (6.7 g/dL). The best leukocyte differential cell count was obtained at A2B1 (lymphocytes: 89%; monocytes: 23,33%; basophils: 0.10%; neutrophils: 22%). The conclusion of this study is the administration of recombinant Chlorella vulgaris nanovaccines based on silver nanoparticles C-AgNPs can be used in the prevention of Viral Nervous Necrosis virus infection and the best dose of nanovaccine administration is 33 μl which is an effective vaccine immunogenicity level in improving the immune system of (Epinephelus sp.) seen from the blood cellular response of (Epinephelus sp.). Further research is needed on the effect of recombinant nanovaccines based on C-AgNPs nanoparticles directly on grouper in floating net cages or aquaculture ponds to control pathogens by increasing the fish immune system, namely white blood cells (leukocytes) and differential leukocytes.
