Abstract
Kegiatan budidaya perikanan atau biasa disebut akuakultur pada zaman modern ini berkembang dengan sangat pesat. Perkembangan kegiatan akuakultur ini tentunya berbanding Iurus dengan permasalahan yang dihadapi. Salah satu masalah dalam kegiatan budidaya perikanan adalah serangan hama dan penyakit pada organisme budidaya seperti penyakit vibriosis yang disebabkan oleh bakteri V. alginolyticus. Pengobatan penyakit vibriosis selama ini umumnya menggunakan antibiotik komersial. Sehubungan dengan permasalahan tersebut maka perlu dicarikan bahan alternatif yang dapat mengendalikan bakteri V alginolyticus serta tentunya aman bagi organisme budidaya dan manusia, Salah satu alternatif yang dapat digunakan untuk mengatasi serangan bakterial adalah dengan menggunakan antimikroba yang berasal dari bahan alami tumbuhan seperti daun sukun (A. altilis). Penelitian dilaksanakan pada bulan Desember 2022 — Februari 2023 yang bertempat di Laboratorium Eksplorasi Sumberdaya Perikanan dan Kelautan dan di Laboratorium Budidaya lkan Divisi Parasit dan Penyakit lkan. Metode yang digunakan adalah metode kuantitatif eksperimental. Pengambilan data pada penelitian ini menggunakan metode observasi secara Iangsung. Rancangan penelitian yang digunakan adalah RAL (Rancangan Acak Lengkap). Berdasarkan uji pendahuluan didapakan 5 dosis perlakuan di antaranyaA(15 ppm), B (30 ppm), C (45 ppm), D (60 ppm), dan E (75 ppm) dengan masing-masing 3 kali ulangan, ditambahkan dengan variabel kontrol berupa kontrol positif antibiotik ciprofloxacin dan kontrol negatif akuades. Pengujian aktivitas antibakteri dilakukan dengan metode difusi cakram untuk melihat pengaruh perlakuan dosis yang digunakan terhadap zona bening atau zona hambat yang dihasilkan. Hasil penelitian yang berjudul aktivitasantibakteri ekstrak kasar daun sukun (A. altilis) terhadap bakteri V alginolyticus secara In vitro dapat diambil kesimpulan bahwa ekstrak daun sukun memiliki pengaruh terhadap pertumbuhan bakteri V alginolyticus secara In vitro yang bersifat membunuh atau bakteriosidal dengan zona hambat tertinggi adalah perlakuan E (75 ppm) pada ulangan 3 yaitu 8,34 mm dan zona hambat terendah adalah perlakuan A (1 5 ppm) pada ulangan 2 yaitu 6,82 mm. Hubungan antara dosis perlakuan dengan rata-rata diameter zona bening menghasilkan grafik persamaan kurva y = 0,0227x + 6,5768 dan koefisien determinasi R’ = 0,9836. Berdasarkan kurva yang dihasilkan menunjukkan bahwa semakin tinggi dosis yang diberikan, zona bening yang terbentuk akan semakin besar. Berdasarkan hasil pengukuran zona bening, ekstrak kasar daun sukun (A. altilis) tergolong dalam kategori daya hambat sedang dikarenakan daya hambatnya berada dibawah 10 mm.
English Abstract
Aquaculture activities or commonly called aquaculture in modern times are developing very rapidly. The development of aquaculture activities is of course directly proportional to the problems faced. One of the problems in aquaculture activities is the attack of pests and diseases on cultivated organisms such as diseases vibriosis caused by bacteria V. alginolyticus. Disease treatment vibriosis has generally used commercial antibiotics. In connection with these problems it is necessary to look for alternative materials that can control bacterial/. alginolyticus and of course safe for cultivated organisms and humans. One alternative that can be used to overcome bacterial attacks is to use antimicrobials derived from natural plant ingredients such as breadfruit leaves (A. altilis). The research was conducted in December 2022 — February 2023 which took place at the Marine and Fishery Resources Exploration Laboratory and at the Fish Aquaculture Laboratory, Division of Fish Parasites and Diseases. The method used is a quantitative experimental method. Collecting data in this study using a direct observation method. The research design used was RAL (Completely Randomized Design). Based on the preliminary test, there were 5 treatment doses including A (15 ppm), B (30 ppm), C (45 ppm), D (60 ppm), and E (75 ppm) with 3 repetitions each, added to the control variable in the form of positive control of ciprofloxacin antibiotic and negatif control of distilled water. Antibacterial activity testing was carried out using the disc diffusion method to see the effect of the dose treatment used on the resulting clear zone of inhibition zone. The results of the study entitled antibacterial activity of the crude extract of breadfruit leaves (A. altilis) against bacteriaV. alginolyticus regularlyln vitro it can be concluded that breadfruit leaf extract has an influence on bacterial growth V. alginolyticus regularlyln vitro Which toThe killing or bactericidal properties with the highest inhibition zone was treatment E (75 ppm) in repetition 3, namely 8.34 mm and the lowest inhibition zone was treatment A (15 ppm) in repetition 2, namely 6.82 mm. The relationship between the treatment dose and the average diameter of the clear zone produces a graph of the linear curve equation y = 0,0227x + 6,5768 and the coefficient R’ = 0,9836. Based on the resulting curve, it shows that the higher the dose given, the larger the clear zone formed. Based on the results of clear zone measurements, the crude extract of breadfruit leaves (A. a/t/Iis) belongs to the category of moderate inhibition because its inhibition is below 10 mm.
