Abstract
Infeksi malaria dapat menyebabkan hemolisis pada eritrosit yang terinfeksi maupun tidak terinfeksi serta diperparah dengan adanya diseritropoiesis sumsum tulang sehingga menyebabkan abnormalitas pada profil sel darah merah. Tablet ekstrak kayu songga (Strychnos lucida R. Br.) pada penelitian sebelumnya menunjukkan aktivitas antimalaria. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian tablet ekstrak kayu songga terhadap profil sel darah merah mencit model malaria terinfeksi Plasmodium berghei ANKA-sensitif klorokuin yang meliputi RBC, hematokrit, hemoglobin, MCV, MCH, dan MCHC. Pemeriksaan profil sel darah merah dilakukan menggunakan Hematology Analyzer Rayto RT-7600 for Vet ®. Mencit yang terinfeksi malaria diberi tablet ekstrak kayu songga dosis 100, 300, dan 500 mg/kgBB lalu dibandingkan dengan kelompok mencit yang diberi klorokuin 25 mg/kgBB. Hasil menunjukkan bahwa semua kelompok uji mengalami anemia makrositik hipokromik yang dapat dilihat dari kadar RBC, hematokrit, hemoglobin, MCHC di bawah kadar normal standar, MCV di atas kadar normal standar, dan MCH normal. Uji korelasi statistik menunjukkan pola semakin tinggi dosis tablet ekstrak kayu songga semakin tinggi pula kadar RBC, hematokrit, dan hemoglobin dengan korelasi lemah dan tidak signifikan pada RBC dan hematokrit (r=0,377;p=0,183 dan r=0,320;p=0,265), serta korelasi sedang dan tidak signifikan pada hemoglobin (r=0,451;p=0,105). Uji komparasi statistik tablet ekstrak kayu songga tidak menunjukkan perbedaan yang signifikan dibandingkan dengan klorokuin (p>0,05) pada profil sel darah merah kecuali pada kadar MCV (p<0,05). Kadar MCV kelompok tablet ekstrak kayu songga dosis 100 mg/kgBB (60,34±5,47fL) lebih tinggi daripada kelompok klorokuin (52,04±2,17fL) yang lebih mendekati nilai normal (46,3-50,3fL).
English Abstract
Malaria causes hemolysis in infected and uninfected erythrocytes. Bone marrow dyserythropoiesis could also exacerbated this condition thus causes abnormality in red blood cells (RBC) profile. Tablet formulation of Songga branch stem extract (Strychnos lucida R.Br.) shows antimalarial activity on the previous research. This study aims to determine the effect of songga branch stem extract tablets (SET) on RBC profile of malaria model mice infected with Plasmodium berghei ANKA-chloroquine sensitive which includes RBC, hematocrit, hemoglobin, MCV, MCH, and MCHC. RBC profile was examined by Hematology Analyzer Rayto RT-7600 for Vet®. Malaria-infected mice were given SET at 100, 300, and 500mg/kgBW dose then compared to 25 mg/kgBW chloroquine group. The RBC, hematocrit, hemoglobin, and MCHC levels that less than normal levels along with exceeding level of MCV indicate that all groups had macrocytic hypochromic anemia. Statistical correlation test shows that a higher dose of SET is directly proportional to RBC, hematocrit, and hemoglobin levels but with weak and not significant degree between RBC and hematocrit (r=0.377;p=0.183 and r=0.320;p=0.265). This also applied to hemoglobin but with moderate and no significant correlation (r=0.451;p=0.105). Statistical comparison test of SET compared to chloroquine group did not show a significant difference (p>0.05) in the red blood cell profile except for MCV levels (p<0.05). The MCV level of the SET group at a dose of 100 mg/kg BW (60.34 ± 5.47fL) was higher than chloroquine group (52.04 ± 2.17fL) which was closer to the normal value (46.3-50.3fL).
