Perbedaan Antara Ekspresi Cytokeratin 12 Dan Cytokeratin 13 Pada Kornea Tikus Model Limbal Stem Cell Deficiency

Abstract

Tujuan : Mengetahui perbedaan ekspresi cytokeratin 12 dan cytokeratin 13 pada kornea tikus model Limbal Stem Cell Deficiency. Metode : Penelitian ini merupakan true experimental in vivo pada kornea tikus model Limbal Stem Cell Deficiency yang dipapar dengan menggunakan benzalkonium klorida (BAK) 0.5% sebanyak empat kali sehari selama 30 hari. Pada akhir penelitian dilakukan pemeriksaan dengan menggunakan slitlamp biomikroskop (Inami L-0189) dan pewarnaan fluoresen untuk menilai klinis LSCD pada kornea tikus model, kemudian dilakukan enukleasi untuk mengambil jaringan konjungtiva-kornea untuk dilakukan pengamatan morfologi jaringan menggunakan pewarnaan Hematoksilin Eosin (HE), serta dilakukan pemeriksaan double staining imunofluoresen untuk menilai eskpresi cytokeratin 12 (CK 12) dan cytokeratin 13 (CK 13). Hasil : Pemberian BAK 0.5% empat kali sehari selama 30 hari menghasilkan terjadinya LSCD pada kornea tikus model yang ditandai secara klinis dengan adanya neovaskularisasi kornea, defek epitel kornea, dan tes fluoresen positif. Pada pemeriksaan HE didapatkan adanya neovaskularisasi, dan hiperplasia sel keratosit pada lapisan stroma kornea tikus. Penanda epitel konjungtiva CK 13 meningkat pada kornea kelompok perlakuan, sedangkan penanda epitel kornea CK 12 menurun pada kornea kelompok perlakuan. Didapatkan perbedaan yang signifikan ekspresi CK 12 dan CK 13 pada kornea kelompok perlakuan (p=0.000) Selain itu juga didapatkan Hubungan bersifat negatif dan berkekuatan sedang. Semakin tinggi ekspresi CK 13 maka semakin rendah CK 12 pada kornea kelompok perlakuan (p=0.049). Kesimpulan : Terdapat perbedaan ekspresi CK 12 dan CK 13 pada kornea tikus model LSCD.

English Abstract

Methods: This study is a true experimental in vivo on mouse cornea Limbal Stem Cell Deficiency model exposed by using benzalkonium chloride (BAK) 0.5% four times a day for 30 days. At the end of the study, an examination using slitlamp biomicroscope (Inami L-0189) and fluorescent staining to assess the clinical LSCD on the cornea of mouse models, then performed enucleation to take conjunctiva-corneal tissue to observe tissue morphology using hematoxylin eosin (HE) staining, and double immunofluorescent staining to assess the expression of cytokeratin 12 (CK 12) and cytokeratin 13 (CK 13). Results: Administration of 0.5% BAK four times daily for 30 days resulted in the occurrence of LSCD in the cornea of model mouse, clinically characterized by the presence of corneal neovascularization, corneal epithelial defects, and positive fluorescent tests. On HE staining examination was found the presence of neovascularization, and hyperplasia of keratocyte cells in the corneal stromal layer. Conjunctival epithelial marker CK 13 was increased in the cornea of the treatment group, while corneal epithelial marker CK 12 was decreased in the cornea of the treatment group. Significant differences were obtained in the expression of CK 12 and CK 13 in the cornea of the treatment group (p=0.000). In addition, it is also obtained that the relationship is negative and of medium strength. The higher the expression of CK 13, the lower the CK 12 in the cornea of the treatment group (p=0.049). Conclusion : There are differences in the expression of CK 12 and CK 13 in the cornea of mouse lscd model.
